人参皂苷Rb1干预长春新碱抑制骨髓基质细胞增殖的研究

摘　　要：目的:探讨人参皂苷Rb1(GS Rb1)促进小鼠骨髓基质细胞(BMSCs)生长及干预长春新碱(VCR)抑制小鼠BMSCs增殖的作用。方法:正常对照组,GS Rb1(1.0、5.0、10.0、15.0μg/ml)诱导BMSCs增殖组,VCR(2.5、5.0、10.0、15.0μg/ml)抑制BM-SCs增殖组,GS Rb1干预VCR抑制BMSCs增殖组,应用MTT法测定BMSCs增殖。结果:①与正常对照组比较,GS Rb1 1.0～10.0μg/ml随浓度升高其促进BMSCs增殖也增加,GS Rb1为10.0μg/ml时差异具有显著性(P<0.05);VCR 2.5～15.0μg/ml随浓度的升高抑制BMSCs增殖也增强,VCR 5.0～15.0μg/ml时差异具有显著性(P<0.05);GS Rb1 1.0～15μg/ml各组与VCR 5.0～15.0μg/ml各组比较,以及GS Rb1 10.0μg/ml组与VCR 2.5μg/ml组比较其BMSCs增殖显著增加(P均<0.05)。②GS Rb1 5.0μg/ml、10.0μg/ml1、5.0μg/ml分别与VCR 5.0μg/ml培养,其能显著干预VCR对BMSCs增殖的抑制(P均<0.05)。结论:长春新碱明显抑制骨髓基质细胞生长;人参皂苷Rb1可促进骨髓基质细胞生长且具有干预长春新碱对骨髓基质细胞增殖的抑制作用。Objective：To investigate the effect of Ginsenoside Rb1（GS Rb1） on promoting the proliferation of bone marrow stromal cells（BMSCs） and interfering with suppressing proliferation of marrow stromal cells by vincristine（VCR）.Methods：The experimental groupgs consisted of normal control group;ginsenoside Rb1 group（1.0,5.0,10.0,15.0μg/ml）;VCR group（2.5,5.0,10.0,15.0μg / ml）;Ginsenoside Rb1 interfering with VCR group,and MTT determination.Results：The BMSCs proliferation promoting effect was increased with GS Rb1 concentration raising from1.0μg/ml to 10.0μg/ml,and there was statistical significance for OD value comparing with normal control group in GS Rb1 10.0μg/ml（P〈0.05）.The BMSCs proliferation inhibiting effect was increased with VCR concentration raising from 2.5μg/ml to 15.0μg/ml,and there was statistical significance for OD value comparing with normal control group from 5.0μg/ml to 15.0μg/ml（P〈0.05）.The BMSCs proliferation promoting effect was significantly increased in GS Rb1 from 1.0μg/ml to 10.0μg/ml comparing with VCR from 5.0μg/ml to 15.0μg/ml（P〈0.05） and in GS Rb1 10.0μg/ml comparing with VCR 2.5μg/ml（P〈0.05）.The BMSCs suppression was significantly blocked in GS Rb1 from 5.0μg/ml to 15.0μg/ml ＋ VCR 5.0μg/ml comparing with VCR 5.0μg/ml（P〈0.05）.Conclusion：The results suggest that vincristine can inhibit the BMSCs proliferation.Ginsenoside Rb1 can promote the proliferation of BMSCs inhibited by vincristine.
