摘  要:目的:观察番荔枝内酯去乙酰紫玉盘素(Desacetyluvaricin)对人肝癌细胞HepG2中细胞外信号调节激酶(extracellular signal regulated kinase,ERKs)表达水平的影响,探讨去乙酰紫玉盘素对ERK/MAPK信号传导通路的影响及其抗肝癌增殖的机制。方法:以12.52、5、50μg/ml的Desacetyluvaricin处理HepG2细胞,Western Blot检测ERK、p-ERK蛋白表达水平的变化,分析去乙酰紫玉盘素对ERK/MAPK信号通路的影响。结果:Desacetyluvaricin处理HepG2细胞后,ERK和p-ERK蛋白表达水平明显下降,并呈剂量依赖关系,其中Desacetyluvaricin以50μg/ml的浓度处理HepG2细胞时免疫印迹条带的发光强度最弱,ERK和p-ERK蛋白的表达水平最低。结论:去乙酰紫玉盘素抗肝癌增殖可能与其抑制ERK/MAPK通路有关。Objective:To observe the effect of desacetyluvaricin on extracellular signal regulated kinase(ERKs) expression in hepatoma carcinoma HepG2 cells,and explore the mechanisms of the antitumor effects of desacetyluvaricin.Methods:HepG2 cells were treated with desacetyluvaricin,Western Blot was performed to examine the expressions of ERK and p-ERK in the cells.Results :The expression of ERK and p-ERK were significantly lowered in the cells after treatment.Conclusion :desacetyluvaricin can resist proliferation of hepatoma cells,and the possible mechanisms may be related to the inhibition of ERK/MAPK pathway in HepG2 cells.
