摘 要:目的:观察两种造模方法对大鼠实验性肾病模型的肾功能及病理变化的影响,为建立适宜的阿霉素致大鼠肾病模型提供参考。方法:采用二次尾静脉注射阿霉素(ADR)法及单侧肾摘除加重复尾静脉注射ADR法造成大鼠实验性肾病模型,测定各组肾功能(尿素氮、肌酐)及观察各组病理改变。结果:二次尾静脉注射组和单侧肾摘除加重复尾静脉注射组的大鼠均有肾功能和病理改变,①4周末和10周末时,两组肌酐浓度与造模前比均显著升高(P<0.05);4周末时,两组尿素氮浓度均升高,单侧肾摘除加重复尾静脉注射组在10周末时尿素氮浓度亦能极显著升高(P<0.01)。②光镜下两组大鼠均有肾间质灶性炎症病理改变,肾小管及肾小球受损。单侧肾摘除加重复尾静脉注射组较二次尾静脉注射组在受损程度及灶性病变程度和范围上明显加重。结论:这两种造模方法所致的大鼠肾功能及病理变化不一,单侧肾摘除加重复尾静脉注射ADR法造成的肾病模型较之二次尾静脉注射ADR法,肾功能及病理改变更为典型稳定,效果更佳。Objective:To investigate the effects of two different methods on rat experimental nephropathy model with the pathological and renal function changes,to provide reference for building the suitable adriamycin(ADR)-induced nephropathy model.Methods :The rat experimental nephropathy model was established by repeated injecting ADR and unilateral nephrectomy combined repeated injecting ADR,to test the renal function(Bun,Cr) and to obeserve the pathological changes of the two groups.Results:The renal function and pathological changes of rats had changes in repeated injecting ADR group and unilateral nephrectomy combined repeated injecting ADR group.①In 4th and 10th weekends,The concentration of Cr in both model groups were increased significantly compared with before;In 4th weekend,the concentration of Bun in both groups were increased,Moreover,The concentration of Bun in the unilateral nephrectomy combined repeated injecting ADR group was increased very significantly in 10th weekend.②Under the light microscope,renal interstitial inflammation revealed focally in both model groups,combined with renal tubular and glomerular injury.Moreover,The unilateral nephrectomy combined repeated injecting ADR group had the more severe condition in the injury and pathological changes than the repeated injecting ADR group.Conclusion:The renal function and pathological changes in the two experimental models are different.The unilateral nephrectomy combined repeated injecting ADR method induces an optimal outcome,with the more typical and stable changes than the repeated injecting ADR method.
