^(147)Pm诱发人白血病细胞凋亡的电镜形态及DNA凝胶电泳研究

摘　　要：研究了受荧光涂料激发能源１４７Ｐｍ 辐照人白血病ＨＬ－ ６０ 细胞所诱发的细胞凋亡。实验中估算了１４７Ｐｍ 在不同阶段培养细胞中的辐射累积吸收剂量。通过透射电镜的形态观察，发现当ＨＬ－６０ 细胞在受１４７Ｐｍ 辐照时，可诱发明显的核断裂、核边聚，以及呈现膜包裹着的凋亡小体形成为特征的细胞凋亡。进而抽提了ＨＬ－ ６０ 细胞的ＤＮＡ，进行琼脂糖凝胶电泳观察，显示出阶梯状条带形成的细胞凋亡特征。从而提示了１４７Ｐｍ 辐照可导致人白血病ＨＬ－ ６０ 细胞凋亡。At present,the apoptosis in human leukemia HL-60 cells was studied after irradiation with 147 Pm.The cumulative radiation absorption dose of 147 Pm in cultural cells through different periods was estimated.The morphological changes observed under electron microscope indicated that HL-60 cells after exposure to 147 Pm displayed nuclear fragmentation and nuclear margination,as well as the membrane bounded apoptotic body formation.The DNA agarose gel electrophoretic observations showed the DNA ladder pattern formation in HL-60 cells.The experimental results showed that apoptosis induced by 147 Pm in HL-60 cells was dependent on the 147 Pm treated time.
