摘 要:目的 :研究骨形态发生蛋白 (BMP)及骨祖细胞的分布与作用 ,探讨不同骨愈合方式的细胞与分子机理。方法 :制造兔桡骨中下段骨折 ,缺损 3mm ,不同时期取骨痂作HE及BMP免疫组化染色 ,显微镜下观察结果并作计算机图像分析。结果 :膜内成骨区及软骨内成骨区骨祖细胞、成骨细胞、成软骨细胞及未成熟的骨细胞BMP染色呈强阳性 ;但肥大的软骨细胞和成纤维细胞为阴性染色。计算机图像分析显示 :膜内成骨区较软骨内成骨区有较高BMP浓度及较多阳性着色细胞。结论 :骨愈合方式与局部BMP浓度及骨祖细胞的来源与分布有关。Objective:To study the distribution of BMP and osteoprogenitor cells during fracture healing and to understand the cellular and molecular mechanism.Methods:3 mm bone defects were made in the middle of radius in rabbits and the callus was obtained on different days after operation.Utilizing immunohistochemical staining and HE staining,we observed the result with microscope and analyzed it by computer imaging system.Results:During fracture healing,there are two types of callus,one forming bone directly and the other forming cartilage.The osteoprogenitors in periosteum and bone marrow differentiate to osteoblasts and osteocytes,directly produce membranous bone.Otherwise,the interfragmentary callus undergo subchondral bone formation.The result of immunostaining show that BMP is distributed in osteoprogenitors,osteoblasts,chondroblasts and immature osteocytes,whereas chondrocytes and fibroblasts are negative.Strong staining is also seen in marrow and muscle near the bone defects.Analyzing with computer imaging system,we found that there were higher gradient distribution of BMP and more immunostaining cells in periosteal callus than those in interfragmentary callus.Conclusion:The distribution of BMP and osteoprogenitor cells is related to the type of fracture healing.
