抗纤益心方对扩张型心肌病心力衰竭大鼠心肌基质金属蛋白酶-1及其抑制剂表达的影响

摘　　要：目的观察抗纤益心方对扩张型心肌病心力衰竭大鼠心肌基质金属蛋白酶-1(MMP-1)及其抑制剂(TIMP-1)表达的影响。方法将100只Wistar雄性大鼠随机分为正常组10只、模型组90只,采用自主饮用呋喃唑酮水溶液建立动物模型。8周后造模成功70只,随机分为模型组、卡托普利组、抗纤益心方高剂量组、抗纤益心方低剂量组、中西药联用组,每组14只,每日分别灌胃生理盐水、卡托普利10.125mg/kg、抗纤益心方18.8g/kg、抗纤益心方4.7g/kg、抗纤益心方高剂量和卡托普利混合溶液,干预8周。每组大鼠随机选取6只进行免疫组化法染色,检测心肌基质MMP-1及TIMP-1的表达。结果与模型组比较,中西药联用组、抗纤益心方高剂量组和卡托普利组均明显下调MMP-1表达(P<0.05),上调TIMP-1表达(P<0.05);抗纤益心方低剂量组能下调MMP-1表达(P<0.05),但TIMP-1表达与模型组差异无统计学意义(P>0.05)。结论抗纤益心方能够下调与心肌纤维化有关的MMP-1的表达,并上调TIMP-1的表达,可能是通过减轻扩张型心肌病心力衰竭大鼠心肌组织纤维化,从而抑制心室重构的作用机制之一。Objective To observe the effect of Kangxian Yixin Formula（Formula for relieving fibrosis and tonifying the heart） on the expression of myocardial matrix metalloproteinase-1（MMP-1） and tissue inhibitor of metalloproteinase-1（TIMP-1）.Methods One hundred Wistar male rats were randomized into normal group,10 rats,and model group,90 rats.The dilated cardiomyopathy models were established by raising with furazolidone solution for 8 weeks.Then 70 successfully made models were randomized into 5 groups,i.e.model group,captopril group,Kangxian Yixin Formula high-dose and low-dose groups,and integrated Chinese and western medicine group,with 14 in each.The models were given normal saline,captopril 10.125mg/kg,Kangxian Yixin Formula 18.8g/kg or 4.7 g/kg,and the mixture of Kangxian Yixin Formula high-dose and captopril respectively as intervention for 8 weeks.Six rats of each group randomly selected were treated with immunohistochemical staining for measuring the expressions of MMP-1 and TIMP-1.Results Compared with the model group,the MMP-1 expression in the integrated Chinese and western medicine group,Kangxian Yixin Formula high-dose group,and captopril group was significantly down-regulated（P〈0.05）,the TIMP-1 expression up-regulated（P〈0.05）.MMP-1 expression in the Kangxian Yixin Formula low-dose group was down-regulated（P〈0.05）,while its TIMP-1 expression was not different from that in the model group（P〉0.05）.Conclusion Kangxian Yixin Formula could effectively down-regulating MMP-1 expression and up-regulating TIMP-1 expression.This may be one of its functional mechanisms in inhibiting the remodeling of ventricle of heart through relieving the myocardial fibrosis of dilated cardiomyopathy heart failure rats.
