摘 要:采用荧光光谱法,在pH=7.4的磷酸盐缓冲溶液(PBS)中研究了染料木苷(Genistin,Ge)与牛血清白蛋白(BSA)的作用方式及作用机理。结果表明,Ge可静态猝灭BSA的内源荧光,二者之间的结合位点数为1.4,温度对结合常数影响不大,300K、306K、310K和313K的结合常数平均为4.31×104 L.mol-1。热力学参数表明二者之间的作用力主要是静电力。同步荧光光谱表明Ge使色氨酸残基的微环境亲水性增强。本研究对于阐明Ge与蛋白的相互作用机理具有借鉴意义。The interaction mechanism and mode of genistin(Ge) with bovine serum albumin(BSA) in pH 7.4 PBS were studied by fluorescence spectroscopy.The results showed that the intrinsic fluorescence of BSA could be quenched by Ge.Furthermore,the number of binding site of 1.4 was obtained.And the average apparent binding constant at 300 K,306 K,310 K and 313 K was 4.31×104 L/5mol-1.The thermodynamic parameters determined by van't Hoff analysis indicated that the type of force was electrostatic force.Synchronous fluorescence spectra showed that the microenvironment of tryptophan residues was more hydrophilic when Ge was added.So,this study could be significant for understanding the interaction mechanism between Ge and proteins.
