摘 要:目的:应用免疫组化法观察蛋白激酶C(PKC)及热休克蛋白70(HSP70)表达,探讨参元丹的药理性预适应作用及其机制。方法:选取Wistar大鼠60只,随机分为6组,分别造模、取血、取心肌组织。应用HE染色方法对组织切片进行染色,4倍镜下采用彩色病理图像分析系统分析心肌梗死面积。应用免疫组化试剂盒对组织切片进行处理。20倍镜下观察PKC及HSP70表达情况。结果:彩色病理图像分析参元丹能够代替心绞痛的缺血、缺氧的刺激,减小心肌梗死面积,具有药理性预适应的心脏保护作用。PKC的表达情况:假性缺血预处理组的心肌组织中偶见微弱的一处表达。缺血预处理组和药物预处理+缺血预处理组的大鼠心肌组织中PKC大量的表达,药物预处理组大鼠心肌组织的PKC表达充分,提示PKC在参元丹的药理性预适应样心肌保护过程中起作用。HSP70的表达情况:对照组、再灌注组、假性缺血预处理组、缺血预处理组HSP70没有表达,药物预处理组HSP70有表达,药物预处理+缺血预处理组HSP70表达明显,提示HSP70的表达与参元丹的作用相关。结论:参元丹具有药理预适应样作用;参元丹预适应心肌保护作用的机制可能与PKC和HSP70的介导有关。Objective:Applying immunohistochemistry method to observe protein kinase C(PKC) and hot shock protein(HSP70) expression to discusse pharmacology preconditioning function and mechanism of Shenyuandan(SYD) which can supply the vital energy and promote the blood circulation.Methods:Select 60 clean healthy adult Wistar rats and randomly divided them into 6 groups.Then established model,drawn blood,and took the cardiac muscle organization separately.In the end we used the HE staining method to dyeing the tissue slice,put them under 4 times of mirrors,used the colored pathological image analysis system to analysis the cardiac arrest area.Then we used HSP70 immunohistochemical tissue kit to dealing with the tissue.Under 20 times of mirrors observed PKC and the HSP70 expression situation.Results:Colored pathology image analysis showed that SYD could replace the angina pectoris to lack the blood and the oxygen deficit stimulation,and could reduce the cardiac arrest area,which proved it had a pharmacological preconditioning of the heart protective effect.PKC expression situation:Pseudo-myocardial ischemic preconditioning group in the cardiac muscle organization the partner to see a weak expression.In the ischemic preconditioning group and drug pretreatment + ischemic preconditioning group the PKC's expression of rat cardiac tissue was fully,PKC expressed fully in rat cardiac tissue of the drug pretreatment group,which suggested that PKC had a protective effect in the SYD pharmacology pre-adaptation type on cardiac muscle protection process.HSP70 expression situation:In the control group,reperfusion group,pseudo-ischemic preconditioning group,ischemic preconditioning group there were no expression of HSP70,in the drug pretreatment group HSP70 had the expression and in drug pretreatment + ischemic preconditioning group HSP70 had expressed obviously.The resuts showed that the expression of HSP70 correlated to SYD-related effect.Conclusion:SYD had a pharmacological preconditioning-like effect.SYD
