补肾健脾方对骨质疏松骨骼肌线粒体氧化应激的影响

摘　　要：目的:探讨补肾健脾方对骨质疏松骨骼肌线粒体氧化应激的调控作用。方法:6月龄的雌性SD大鼠随机分为模型组、雌激素组、中药组和空白对照组。治疗3个月后,收集肌肉组织进行线粒体通透转换孔和细胞色素C氧化酶的活性检测。结果:在线粒体通透转换孔活性比较方面,雌激素组、空白对照组与模型组比较,差异均有统计学意义(P<0.05);在细胞色素C氧化酶比较方面,空白对照组与模型组比较,差异有统计学意义(P<0.05)。中药组与模型组在上述两组比较,差异均无统计学意义。结论:雌激素表现出对线粒体氧化应激功能的调控能力,因此认为以骨骼肌作为药物靶点治疗骨质疏松症是可能实现的。而中药尽管在实验中均没有出现阳性结果,但数值表明仍值得我们进一步研究。Objective：To study the effect of invigorating kidney and strengthening spleen prescription on the oxidative stress of skeletal muscle mitochondrion in osteoporosis.Methods：Six-month-old female rats were divided into 4 groups：normal control,ovariectomized,estrogen supplemented and traditional Chinese herbs.After 3 months of treating,mitochondrion were extracted from the muscle tissue that was collected during the operation,and cytochrome coxidase（COX）,the mitochondrial permeability transition pore（MPPT） were detected.Results：In the MPPT,there was a statistically significant difference that the ratio of OD value of normal control group and of the estrogen supplemented group respectively compared with the ovariectomized group（P0.05）.In the COX,there was a statistically significant difference between the normal control group and the ovariectomized（P0.05）.There were no a statistically significant difference that the value of the ovariectomized group and of the traditional Chinese herbs group in the MPPT and COX.Conclusion：Estrogen could regulate the oxidative stress of skeletal muscle mitochondrion in osteoporosis,so skeletal muscle as drug targets to treat osteoporosis was possible.Although the invigorating kidney and strengthening spleen prescription had not positive results,the values indicated that the Chinese herbs in the regulation of mitochondrial oxidative stress had a broad prospect.
