摘  要:目的:通过组分填充模拟炮制方法,探讨大黄饮片炮制前后主要成分的变化规律。方法:以"空白饮片"为载体,填充不同组分进行生大黄、酒大黄、醋大黄、熟大黄和大黄炭饮片的模拟炮制,采用HPLC法分析比较不同组分模拟大黄饮片的主成分变化规律。结果:炮制以后,大黄中的苷类成分主要表现为自身的破坏及向其相应苷元转化,同时其相应的苷元也存在着自身破坏与相应苷类成分向其转化的动态过程。由于部分苷类成分中含有没食子酯基,因此炮制过程中,不仅可转化为相应的苷元,还分解产生没食子酸。另外,鞣质类成分炮制后,一部分被完全破坏,另一部分转化为儿茶素、没食子酸等单体鞣质。这也是大黄熟片和炭片中没食子酸含量显著增加的原因。结论:以空白饮片为载体的组分填充模拟炮制方法与其他模拟炮制方法相比,可以较为真实地反映化学成分在炮制过程中的变化情况,是中药炮制原理研究的一种有效的新方法。Objective:To research on the variation laws of main constituents in the pieces of rhubarb by the simulation processing method.Methods:Taken the 'blank pieces' as the carrier and filled with different components from rhubarb,then processed as five kinds of pieces.HPLC chromatography were used to analysis the changes of main components.Results:After processing,the glycosides were destroyed and converted to its corresponding aglycone,while the corresponding aglycones there were also self-destruction and increased from their corresponding glycosides.At the same time,the glycosides which contained gallic esters not only can be converted to the corresponding aglycones,but also decomposed to gallic acid.In addition,a part of tannins were transformed as catechins and gallic acid.This was the main reasons for the sharply increases of gallic acid in the charred and steaming pieces.Conclusion:Compared with other simulation processing method,our method may reflect the changes of main constituents in different pieces more veritably.It was a more effectively new method for processing principle research.
