摘  要:目的:研究中药丹参对大鼠粘连成纤维细胞增殖与胶原合成的影响。方法:利用原代培养获得大鼠粘连组织成纤维细胞(AFB),以地塞米松为阳性对照药,考察丹参对AFB增殖活性与培养液羟脯氨酸含量(HyP)的影响。结果:随着剂量增加,丹参组AFB活性降低趋势大于地塞米松组,丹参组的IC50也显著低于地塞米松组(P<0.01);丹参各剂量组与地塞米松组的HyP水平均低于空白对照组,存在显著性差异(P<0.01);丹参各剂量组与地塞米松组的HyP水平无显著性差异。结论:抑制AFB增殖,减少其胶原合成是丹参防治术后粘连的作用机制。Objective:To study the effect of Danshen on proliferation and collagen synthesis of cultured fibroblasts from rat adhesions.Method:Adhesive fibroblasts(AFB) of rats were harvested by primary culture.Dexamethasone was used as positive control.Proliferation of AFB and hydroxyproline(HyP) content in culture fluid were detected.Result:Following the increase of drug dosage,AFB viability decreasing tendency of Danshen groups was higher than dexamethasone group.The IC50 of Danshen on AFB was significantly lower than dexamethasone(P0.01);HyP level of Danshen groups and dexamethasone group were significantly lower than blank control(P0.01).But there was no significant difference between Danshen groups and dexamethasone group.Conclusion:To inhibit the proliferation and decrease collagen synthesis of AFB was the preventive mechanism of Danshen on post-surgical adhesion.
