温补肾阳汤对哮喘小鼠Eotaxin表达的干预研究

摘　　要：目的:观察温补肾阳汤对哮喘小鼠外周血、肺组织Eotaxin表达的影响,探讨其抑制气道炎症作用。方法:健康雌性BALB/c小鼠50只,随机分为空白对照组、模型组、醋酸泼尼松组、温补肾阳汤组、温补肾阳汤+醋酸泼尼松组,每组10只。卵白蛋白(OVA)致敏及激发造模方法建立动物模型。计数肺泡灌洗液EOS;光镜下观察肺组织病理学改变;ELISA检测外周血Eotaxin表达;免疫组织化学检测肺组织Eotaxin蛋白水平。结果:药物干预组小鼠外周血、肺组织Eotaxin表达较模型组明显降低(P<0.01),肺泡灌洗液EOS计数明显减少(P<0.01),其中温补肾阳汤+醋酸泼尼松组较温补肾阳汤组外周血、肺组织Eotaxin表达及肺泡灌洗液EOS计数均降低(P<0.05),但与醋酸泼尼松组无差异(P>0.05)。结论:温补肾阳汤可能通过降低哮喘小鼠体内Eotaxin水平,减轻炎症细胞在肺组织局部浸润,从而抑制气道炎症。温补肾阳汤与醋酸泼尼松有一定协同作用,可减轻或避免大剂量糖皮质激素引起的不良反应。Objective：Observing the influence of WTKYD on the expression of Eotaxin in asth matic mice to explore its possible effect in inhibiting airway inflammation.Methods：50 healthy female BALB/c mice were randomly divided into five groups：control group,model group,prednisone group,WTKYD group,WTKYD plue prednisone group（n=10 for each group）.By the classic way of OVA Sensitization and excitation to establish the models.To caculate Eosinophils（EOS） counts of bronchoalveolar lavage fluid（BALF）,to observe pathological change of lung tissue,to detect the expression of Eotaxin in peripheral blood by ELISA and the level of Eotaxin in lung tissue by immunohistochemistry.Results：The expression of Eotaxin in peripheral blood and lung tissue,EOS counts of BALF in three treatment groups were decreased obvuiously compared with model group（P0.01）.And the expression of Eotaxin in peripheral blood and lung tissue,EOS counts of BALF in WTKYD plus prednisone group were reduced dramactically compared with WTKYD group（P0.05）,but there was no statistical difference between WTKYD plus prednisone group and prednisone group（P0.05）.Conclusions：WTKYD can decrease the level of Eotaxin in asthmatic mice and alleviate inflammatory cells infiltration in lung issue,then inhibit airway inflammation.There was synergy between WTKYD and prednisone,a combination can relieve or avoid side effects of high-dose glucocorticoids.
