大黄虫丸抑制小鼠Lewis肺癌及其机理的研究

摘　　要：目的:通过大黄虫丸对荷Lewis肺癌C57BL小鼠瘤体大小、生存期及环氧化酶2(cyclooxygenase,COX-2)表达影响的观察,初步探讨其抗肿瘤的可能机制。方法:造模后次日先随机分成5组,每组10只。据预试验结果确定高(H)、中(M)、低(L)剂量分别为0.18g/mL、0.09g/mL、0.045g/mL,以每只0.2mL灌胃10天;空白组小鼠以NS0.2mL灌胃10天;化疗组以50mg/kg CTX于第1天予腹腔给药。第11天处死小鼠,剖取完整肿块称重并计算抑瘤率。肿瘤组织以免疫组化、RT-PCR及western blot法检测COX-2的表达量。结果:①实验组高(H)、中(M)、低(L)和化疗组抑瘤率分别为29.70%、38.79%、17.58%、63.64%。实验高(H)、中(M)组与空白组比有统计学意义(P<0.05),低(L)与空白组比无统计学意义(P>0.05)。②实验组(M)、化疗组生存期分别为(19.14±0.77)、(19.80±0.75)天,显著长于空白组的(14.50±0.76)天(P<0.05);生命延长率分别为32.00%、36.56%,但实验组(M)与化疗组在生命延长作用方面无显著差异(P>0.05)。③免疫组化、RT-PCR及western-blot检测显示:大黄虫丸具有下调荷Lewis肺癌小鼠COX-2蛋白表达的作用,以实验组(M)作用最明显。结论:大黄虫丸对荷Lewis肺癌小鼠肿瘤具有明显的抑制作用并延长荷瘤小鼠生存期,作用机制可能与下调COX-2蛋白表达有关。Objective：By observing effects of Dahuang Zhechong Pills on the tumor size、survival and expression of COX2 in mice with Lewis Lung carcinoma,to discuss its anti-tumor effect and molecular mechanism preliminarily.Methods：The mice were divided randomly into five groups the day after modeling and each group included ten mice.According to the pre-test,high-dose,medium-dose and low-dose were quantitated with 0.18g/mL,0.09g/mL,0.045g/mL.The model group was intragastric administrated with NS 0.2ml for ten days;the chemotherapy group was Intraperitoneal administrated 50mg/kg CTX on the first day.After experiment,all mice were killed,weighted and compared the tumor,calculated tumor growth inhibition rates of each group on the 12th day.Immunohistochemical staining,RT-PCR and western blot were use to check the protein expression of COX-2.Results： ①The inhibition rates in experimental groups of high（H）,medium（M）,low（L） dosages and the chemotherapy group were respectively 29.70 percent,38.79 percent,17.58 percent,63.64 percent.The group（H）,group（M） compared with the chemotherapy group had a significance（P0.05）;The group（L） compared with normal control group had no significance（P0.05）;②Survival in Group（M） and chemotherapy group,which is respectively（19.14±0.77）D,（19.80±0.75）D,is significantly longer than that（14.50±0.76）D in the model group（P0.05）.Life-prolonging rates are 32.00%,36.56%;There was no significant difference between group（M） and chemotherapy group in survive.③The results from Immunohistochemical staining,RT-PCR and western blot showed that the protein expression of COX-2 was reduced,which was obvious in Group（M）.Conclusion： Dahuang Zhechong Pills inhibited Lewis lung carcinoma in C57BL mice and prolonged their lives,Whose anticarcinogenic effect may depend on reduce the protein expression of COX-2.
