摘  要:目的:通过研究黄芪注射液对实验性胰腺纤维化大鼠氧化应激反应的影响,探索其缓解胰腺纤维化的机制。方法:大鼠随机分为6组,模型组和治疗组采用胰胆管注射2%TNBS溶液的方法制备慢性胰腺纤维化大鼠模型,治疗组分别给予低、中、高剂量的黄芪注射液治疗;造模后第6周,ELISA检测血清MDA含量和GSH-PX活性、免疫组化检测胰腺α-SMA和Ⅰ型胶原的含量。结果:与模型组相比,随着黄芪注射液的剂量的增加,除了GSH-PX活力逐渐恢复以外,治疗组大鼠血清中MDA含量逐渐降低且胰腺中α-SMA和Ⅰ型胶原含量逐渐较少,且均具有有统计学意义。结论:黄芪注射液可以减弱CP的氧化应激,抑制PSC活化,缓解胰腺的纤维化。Object: To investigate the effect of Radix Astragali injection on oxidative stress from rat with chronic pancreatic fibrosis,and to explore the mechanism of alleviation pancreatic fibrosis.Method: Rats were randonmly divided into six groups.The injection method of 2% TNBS through rat's pancreatic duct was undertaken to set up the animal model of pancreatitis.The rats which included in low dose group,moderate dose group and high dose group were treated by Radix Astragali injection.At the end of 6th week,all the survived rats were sacrificed.The expressions ofα-SMA and collagenⅠwere detected by the method of immunohistochemistry.The content of MDA and the activity of GSH-PX in serum were detected by the method of ELISA.Result: Both the content of MDA in serum and the content of α-SMA and collagenⅠsecreted by pancreas was dropped respectively by Radix Astragali injection except that activity of GSH-PX showed opposite trend.All effects showed a dose-dependence,comparing with the control group,with significant differences(P0.05).Conclution: Radix Astragali injection can decrease the effect of oxidative stress of CP and inhibit activation of PSC,resulting in alleviating the fibrosis of the pancreas.
