红景天苷对常压肺心病大鼠保护作用的研究

摘　　要：目的:研究红景天苷对常压慢性低O2诱发的肺动脉高压、肺心病的保护作用。方法:将SD大鼠50只随机分为正常对照组、低O2、低O2+红景天苷(低、中、高)组。采用图像分析、免疫组化技术,透射电镜等方法,检测各组大鼠肺动脉平均压(mPAP)、右心指数、肺小动脉显微和超微结构,Ⅰ、Ⅲ型胶原表达的变化。结果:低氧组mPAP、右心指数、肺小动脉显微结构指标:管壁面积/管总面积、肺细小动脉中膜平滑肌细胞核密度、肺细小动脉中膜厚度较对照组明显升高,肺血管重建,肺小动脉Ⅰ型胶原表达增加。红景天苷可减轻上述改变,高剂量组尤为显著;但Ⅲ型胶原各组间无明显差别。结论:红景天苷可降低慢性低O2诱导的肺动脉高压,减轻肺血管结构重建,其机制与抑制肺动脉Ⅰ型前胶原增殖有关。Objective： To study the protective effect and mechanism of Salidroside on cor pulmonale rats induced by chronic hypoxia in normal pressure.Methods： Fifty Sprague-Dawley rats were randomly divided into five groups：normal control group,normal pressure hypoxia group,hypoxia＋Salidroside group（low dose,median dose,high dose）.Pulmonary arterial pressure（mPAP）,Pulmonary arteriole micromorphometric index：WA/TA（vessel well area/total area）、SMC（density of medial smooth muscle cell） and PAMT（media thickness of pulmonary arterioles）,weight ratio of right ventricle to left ventricle plus septum[RV/（LV＋S）],CollagenⅠand Ⅲ were measured.Results： mPAP,RV/（LV＋S）,Ⅰtype collagen,arteriole micromorphometric index were significantly higher in rats of hypoxia group than those of control group.Salidroside could reverse the changes mentioned above.But there was no significant difference in content of Ⅲ type collagen in five groups Conclusions Salidroside can reduce pulmonary hypertension and pulmonary vessel remodeling induced by hypoxia through inhibiting proliferation of collagenⅠ.
