摘  要:四倍体葡萄深受市场的欢迎,但目前葡萄四倍体品种数量较少,遗传背景狭窄。因此,四倍体诱导研究对于葡萄育种和生产都具有重要意义。试验以秋水仙素为诱变剂,采用浸泡单芽茎段法和混合培养法对京秀葡萄进行组培诱导,以探索秋水仙素浓度以及不同处理方法对变异的影响,并利用流式细胞分析仪对诱变效果进行分析。结果表明,对于浸泡单芽茎段法,浓度的影响强于处理时间,最佳诱导浓度及时间为0.3%处理24 h;而对于混培法,处理时间的影响却强于浓度,最佳诱导浓度及时间为2 000 mg.L-1处理30 d。Tetraploid grapes are well received by the market.But now only several tetraploid grape varieties are available in the market and the genetic background is narrow.So tetraploid induction is of great significance for grape breeding and production.In this study,colchicine as mutagen,use soaking single-bud stem and mixed culture,to explore the effects of colchicine concentrations and different treatments on tissue and culture induction of Jingxiu grape.Flow Cytometer was used to identify the effect of induction.The results showed that the effect of concentration was stronger than that of the treat time for soaking single-bud stem,and the best treatment was 0.3% for 24 h;while for mixed culture method,the role of the treat time was stronger than that of concentration,and the best treatment was 2 000 mg·L-1,for 30 d.
