摘 要:目的探索慢性间断性缺氧(CIH)对大鼠额顶部皮质-κB(NF-κB)、细胞间黏附分子-1(ICAM-1)和肿瘤坏死因子-α(TNF-α)表达的影响。方法制作CIH大鼠模型。将30只SD大鼠均分为CIH组(实验组)、空气模拟对照组及空白对照组。采用免疫组织化学方法检测3组大鼠额顶部皮质NF-κBI、CAM-1和TNF-α的表达,并行统计学处理。结果实验组大鼠额顶部皮质NF-κB表达高于空白对照组和空气模拟对照组(均为P<0.05),而空白对照组与空气模拟对照组之间比较,差别无统计学意义(P>0.05);实验组可见ICAM-1在额顶部皮质和基底节区微血管内皮细胞表面表达,另两组未见表达;实验组皮质的TNF-α亦显著高于另两组(P<0.05)。结论 CIH可通过上调大鼠脑皮层NF-κB的表达并促进其目的基因产物ICAM-1和TNF-α的表达介导脑损害。Objective To observe the effect of chronic intermittent hypoxia on NF-κB,ICAM-1,TNF-α expressions in cortex.Methods Thirty male SD rats were randomly divided into three groups: the experimental group,the air mimic group and the blank control group.Immunohistochemistry was used to examine the expression of nuclear transcription factor κB(NF-κB),intercellular adhesion molecule-1(ICAM-1) and tumor necrosis factor-α(TNF-α)in the frontal and parietal cortex cells.Results The expression of NF-κB in cortex cells from the experimental group was significantly higher than those from the air mimic control group(P0.05) and the blank control group(P0.05).The expression of NF-κB in cortex cells of the air mimic control group was not significantly different from those of the blank control group(P0.05).The experimental group showed that ICAM-1 expression in the microvascular endothelial cells,mainly distributed in the frontoparietal cortex and basal ganglia.There was no positive expression of ICAM-1 in blood vessels in the blank control group and the air mimic control group.The expression of TNF-α in cortex cells from the experimental group was significantly increased compared with those from the air mimic control group and the blank control group(P0.05).Conclusion CIH increases the expression of NF-κB and its target gene products ICAM-1 and TNF-α,and therefore affects the cerebral vascular system.
