摘　　要：目的研究Wnt 3α蛋白对脂肪间充质干细胞向成软骨方向分化能力的影响。方法从4 d SD仔鼠腹股沟脂肪垫分离出脂肪组织,通过I型胶原酶消化分离并经免疫荧光鉴定脂肪间充质干细胞。取第三代脂肪间充质干细胞,采用RT-PCR和Western blot方法观察外源Wnt 3α蛋白对脂肪间充质干细胞增殖和向成软骨细胞分化能力的影响。结果在体外分离与培养的脂肪间充质干细胞的培养液中添加Wnt 3α蛋白,明显上调II型胶原和蛋白聚糖的mRNA和蛋白表达水平。结论 Wnt 3α蛋白具有促进大鼠脂肪间充质干细胞增殖和向成软骨细胞分化作用。Objective To investigate the effect of Wnt3α on the chondrocytes differentiation ability of adipose mesenchymal stem cell. Methods The adipose mesenchymal stem cells were isolated from the fat pads of 4 d SD rats with collagenase I. RT - PCR and Western blot were used to observe the effect of Wnt 3α on the chondrocytes differentiation ability of adipose mesenchymal stem cells. Results Wnt 3ct upregulates the expression of collagen II and Proteoglycan. Conclusions Wnt 3α promotes rat adipose mesenchymal stern cells chondrocyte differentiation ability of the adipose mescenchymal stem cells.
