高效液相色谱法测定固本咳喘片中白术内酯Ⅰ和白术内酯Ⅲ的含量

摘　　要：目的建立同时测定固本咳喘片中白术内酯Ⅰ和Ⅲ含量的方法。方法采用AgilentHC-C18柱(4.6mm×250mm,5μm),流动相-乙腈-甲醇-0.2%磷酸溶液(5:52:43),检测波长为222nm,柱温为35℃,流速为1.0mL/m in。结果白术内酯Ⅰ和Ⅲ分别在0.8-15.0μg/mL(r=0.9995)和1.7-33.0μg/mL(r=0.9997)浓度范围内线性关系良好,平均加样回收率分别为98.22%,97.90%;RSD均小于2.0%。结论本方法简便,准确,重复性好,可用于该制剂的质量控制。Objective To establish a method for the determination of atractylenolideⅠand atractylenolide Ⅲ in Guben Kechuan tablets.Method The reversed phase HPLC system consisting the Agilent HC-C18（4.6mm×250mm,5μm）column,the mobile phase consisted of Acetonitrile（5）-Methanol（52）-0.2% H3PO4（43）,the UV detector was set at 222nm,the flow rate was 1.0mL/min,the column temperature was 35℃.Results The linear response range was 0.8～15.0μg/mL（γ=0.9995）and 1.7～33.0μg/mL（γ=0.9997）,respectively.The average recovery of atractylenolideⅠand atractylenolide Ⅲ were 98.22% and 97.90%,respectively,with RSD less than 2.0%.Conclusions The assay is simple,accurate and repeatable,therefore could be used as a reliable tool for the determination of atractylenolideⅠand atractylenolide Ⅲ in Guben Kechuan tablets.
