摘  要:目的分析标本放置温度及时间对中性粒细胞CD64检测结果的影响。方法抽取10例健康体检者EDTA—K2抗凝静脉血,各分装两管,放置室温和4℃保存,用BDFacs Calibur流式细胞仪检测标本采集后30min、6h、12h、24h的中性粒细胞CD64水平。结果随着放置时间的延长,中性粒细胞表达CD64水平有升高的趋势。标本30rain测定CD64结果为(1540.1±1141.0)PE\cell;室温和4℃放置24h后检测CD64结果分别为(1867.8±1186.0)PE\cell和(1730.6±1221.5)PE\ceH,两者结果有明显差异(P〈0.05);室温和4℃放置24h测定CD64结果都与30min测定CD64结果有明显差异(P〈0.05)。结论为确保CD64检测结果准确性,采血后全血标本应在12h内检测,最好放置4℃保存标本。Objective To investigate the influence of storage temperature and duration of specimens on detection of CD64. Methods 10 samples of venous blood anticoagulated with EDTA-K2 were collected. Each sample was placed in two tubes and stored at room temperature and 4℃ separately. The neutrophil CD64 levels in the samples were detected by flow cytometry 30min, 6h, 12h, and 24h after collection. Results The expression level of CD64 in neutrophils increased with time. CD64 was ( 1540.1±1141.0 )PE/cell at 30 min and was ( 1867.8±1186.0 ) PE/cell at room temperature and ( 1730.6±1221.5 ) PE/cell at 4℃ after 24- hour storage, showing a significant difference in the expression level of CD64 between two different temperatures and two sorts of storage duration ( P 〈 0.05 for both comparisons ). Conclusions Blood samples should be stored at 4℃ and detected within 12h after collection to ensure accuracy of the results.
