菊藻丸含药血清对人激素非依赖性前列腺癌细胞增殖和周期的影响

摘　　要：目的探讨含菊藻丸大鼠血清对人激素非依赖性前列腺癌细胞系PC-3增殖和细胞周期的影响。方法以3种不同质量浓度的菊藻丸水剂对成年雄性去势SD大鼠灌胃。另以等量生理盐水灌胃进行对照。末次灌胃后于60～90min时间段内取下腔静脉血制成血清,用以体外培养PC-3;用MTT法测定培养4、24、48、72h的细胞存活率,用流式细胞仪测定培养48h的细胞周期分布。结果高、中剂量的含药血清可抑制PC-3细胞的生长,且抑制作用随时间的延长而增强,低剂量无明显抑制作用。含药血清高剂量组G1-G0期细胞比例低于中、低剂量组,G2/M期细胞比例高于中、低剂量组。结论含菊藻丸血清能抑制PC-3细胞增殖,可用于动物实验进一步明确体内有无抗肿瘤作用;不同质量浓度含药血清对细胞周期影响不同,机制有待研究。Objective：To investigate the effect of the serum containing Juzao Pills on proliferation and cell cycle of hormone-independent prostate cancer.Methods：Adult male castrated SD rats were gavaged with decoction of different concentrations of Juzao pills twice a day for there days.These animals in control group were gavaged with normal saline in the same volume.Between sixty minutes and ninety minutes after the last administration,blood samples were taken from the inferior vena cava of rats.The drug-containing serum and the blank control serum were prepared.PC-3 cells were incubated in the culture medium with serum for 4,24,48 and 72 h,and then cells were measured by MTT assay.Cell cycle was measured by flow cytometry when cells were incubated with the drug-containing serum or blank control serum for 48h.Results：These cell growths were inhibit in the high dose and the middle dose of containing drug serum.The inhibition was enhaced as the action time prolonging.There was no inhibition effect in the low dose group.In the high dose of serum containing drug,the proportion of cells in the G1-G0 was lower than that in the middle dose and the low dose group.Meanwhile,the ratio of G2/M cell in the high dose of serum containing drug was higher than that in the middle dose and the low dose group.Conclusion：The proliferation of PC-3 can be inhibit by serum containing Juzao Pills which could be used in the animal experiment to confirm its anticancer effect in vivo.Different concentration groups have different effects on PC-3 cell cycle.The mechanism might be studied in future.
