摘 要:建立了微流控芯片非接触电导检测法测定片剂中盐酸奈福泮含量的方法。探讨并优化了缓冲溶液种类和配比、添加剂、分离电压和进样时间等电泳分离条件。结果表明,以2 mmol/L HAc+1 mmol/L NaAc(pH4.5)不加添加剂为运行缓冲溶液,分离电压2.00 kV、进样时间10 s时,1 min内可实现盐酸奈福泮快速分离检测。盐酸奈福泮的线性范围为20μg/mL~120μg/mL(r2=0.9992),检出限(LOD)为1.2μg/mL(S/N=3),加标回收率为98.0%~100%,RSD小于2.2%。方法简便快速,重现性好,适用于盐酸奈福泮快速检测和生产质量控制。A new method of microfluidic chip with contactless conductivity detection was established for the determination of nefopam hydrochloride in its tablets.The optimum separation electrophoretic conditions,such as the variety and concentration of buffer solution and additive,separation voltage and injection time etc,were researched.The results indicated that in the optimized running buffer of 2 mmol/L HAc+1 mmol/L NaAc(pH 4.5,no additive),nefopam hydrochloride was well separated and detected in 1 min at the separation voltage of 2.00 kV and injection time 10s.The linear concentration of nefopam hydrochloride ranged from 20 μg/mL to 120 μg/mL(r 2=0.9992),and the limit of detection(LOD)reached 1.2 μg/mL(S/N=3).The recovery of standard addition was 98.0%to 100%and the RSD was less than 2.2%.The method can be used for the rapid detection and the quality control of the product due to its simplicity,rapidity and reproducibility.
