四种基源的川贝母对非小细胞肺癌A549细胞的抑制作用

摘　　要：目的:观察四种不同基源的川贝母(梭沙贝母、暗紫贝母、卷叶贝母、甘肃贝母)对非小细胞肺癌A549细胞的抑制作用。方法:以不同浓度的川贝母作用于A549细胞,分别用MTT法、细胞生长曲线观察四种基源的川贝母对A549增值抑制的影响,及其细胞形态学变化。结果:MTT显示川贝母浓度为10mg/L～150mg/L时,对A549的48、72小时抑制率为5%～92%,呈浓度和时间的依赖关系;梭沙贝母、甘肃贝母、暗紫贝母、卷叶贝母72小时IC50值分别为50mg/L、62mg/L、66mg/L、76mg/L;生长曲线提示:随着川贝母作用时间的延K,细胞增殖抑制率逐渐升高,呈明显的时效和量效关系;荧光倒置显微镜下观察出现明显凋亡形态学的变化。结论:四种不同基源的川贝母都对非小细胞肺癌A549细胞有增值抑制作用,其中梭沙贝母的抑制作用最好,其次分别是甘肃贝母、暗紫贝母、卷叶贝母,但这四种不同基源的川贝母之间作用差异性不大。Objective:To investigate the inhibitory effort of Chuanbeimu(F.delavayi Franch.,F.unibracteata Hsiao et K.C.Hsia,Fritillaria cirrhosa D.Don,F.prezewalskii Maxim.)on A549 cell of human non-small cell lung cancer.Methods:The inhibitory effort of Chuanbeimu on proliferation of A549 cells was evaluated by MTT analysis and growth curve.Their apoptosis inducing effect were demonstrated by morphological observation under fluorescent inverted microscope.Results:After treated with the four different sources of Chuanbeimu for 48 and 72 hours.MTT assay showed the dose range from 10mgL^150mgL had the inhibitory ratio range from 5%~92%.IC50 0f the Chuanbeimu to A549 in 72h were 50mgL,62mgL,66mgL,76mgL respectively;The dose-effect relationship and time-effect relationship were described in the growth curve;The characteristic morphology typical for the four different sources of Chuanbeimu were observed under fluorescent inverted microscope.Conclusions:The four different sources of Chuanbeimu can inhibit the growth of A549 cells in vitro,their mechanism are probably associated with the apoptosis inducement.The inhibitory effect of the F.delavayi Franch.is the best,followed by the F.prezewalskii Maxim.,F.unibracteata Hsiao et K.C.Hsia,Fritillaria cirrhosa D.Don,but there are only few differences between them.
