Galectin-9在小鼠病毒性心肌炎中的表达及意义

摘　　要：目的检测病毒性心肌炎小鼠不同时间点血清中半乳糖凝集素-9（Galectin-9）的水平及心肌组织中Galec-tin-9mRNA的表达,探讨Galectin-9在病毒性心肌炎发生发展中的作用。方法选取58只56周BALB/c小鼠,随机分为病毒组50只和对照组8只。病毒组小鼠腹腔接种0.10mL TC ID50为10-5/L的柯萨奇病毒B3（CVB3）N ancy株病毒液,分别于接种病毒后第7、10、14、28天处死8只小鼠;对照组小鼠腹腔接种0.10mL不含病毒的Eag le氏液,28d时处死小鼠。取心脏,在光镜下观察心肌病理变化,计算心肌组织病理学积分。取外周血,用ELISA检测血清中Galectin-9的含量。采用实时荧光定量PCR（RQ-PCR）检测心肌组织中Galectin-9mRNA的表达。结果病毒组小鼠血清中Galectin-9含量在第7、10、14天均较对照组明显增高（P〈0.01）。病毒组小鼠心肌组织Galectin-9mRNA的表达在第7、10天均较对照组增强（P〈0.01）。病毒组小鼠血清中Galectin-9的含量及心肌组织中Galectin-9mRNA的表达均与心肌组织病理积分呈正相关（r分别为0.75,0.74;P〈0.05）。病毒组小鼠血清中Galectin-9的含量与心肌组织中Galectin-9mRNA的表达呈正相关（r=0.71,P〈0.05）。结论急性期病毒性心肌炎小鼠Galectin-9表达明显升高,且与心肌炎病变严重程度呈正相关,Galectin-9可能在病毒性心肌炎的发生发展中起一定作用。Objective To detect the level of plasma Galectin-9 and expression of Galectin-9 mRNA in the myocardium of viral myocarditis mice at different phases and explore its potential role in the occurrence and development of viral myocarditis.Methods Fifty-eight BALB/c male mice（5 to 6 weeks） were randomly divided into the coxsackie-virus B3（CVB3） group（n=50） and the control group（n=8）.The mice in the CVB3 group were inoculated with 0.10mL TCID50 10-5/L coxsackie-virus B3.Eight mice in CVB3 group were sacrificed on day 7,10,14 and 28 after the inoculation.The mice in the control group were inoculated with 0.10mL Eagle reagent and the mice were sacrificed on day 28.Enzyme-linked immunosorbent assay（ELISA） was used to detect the level of serum Galectin-9.With extracted hearts histopathological changes were observed by light microscope and the myocardial histopathological scores were calculated.Expression of Galectin-9 mRNA of the myocardium was determined by the real-time quantitative polymerase chain reaction（RQ-PCR）method.Result The level of plasma Galectin-9 elevated significantly on day 7,10 and day 14（P〈0.01）.Expression of Galectin-9 mRNA in the myocardium significantly increased on the 7th day and 10th day（P〈0.01）.The level of plasma Galectin-9 and expression of Galectin-9 mRNA were positively correlated with myocardial pathological scores（r=0.75,0.74;P〈0.05）.The level of plasma Galectin-9 in the CVB3 group werepositively correlated with expression of Galectin-9 mRNA（r=0.71;P〈0.05）.Conclusion Expression of Galectin-9 in the viral myocarditis mice were significantly elevated,which was positively correlated with the severity of the myocardium.It may play a role in the occurrence and development of viral myocarditis.
