应用噬菌体随机十二肽库筛选TGF-β1相关模拟肽的实验研究

摘　　要：目的应用噬菌体随机12肽库生物筛选获得转化生长因子-β1(TGF-β1)噬菌体模拟肽。方法以人TGF-β1单克隆抗体为靶,进行4轮生物淘选,随机挑取单克隆噬菌体,ELISA法检测单克隆噬菌体的结合力。对结合力较好的单克隆噬菌体提取噬菌体DNA,进行测序,行序列分析。结果经过4轮生物淘选,特异性噬菌体模拟肽获得了富集,ELISA检测结果显示,获得的一些噬菌体模拟肽具有较好的结合力,测序获得10个与促进成纤维细胞增殖或抑制细胞增殖有关的碱基序列。结论从噬菌体随机十二肽库中可淘选到与TGF-β1相关的噬菌体模拟肽。Objective To harvest TGF-β1 model peptides from a phage display 12-mer peptide library. Methods A phage display 12-mer peptide library was screened 4 times by monoclonal anti-human TGF-β1. Monoclonal phages were randomly selected by ELISA. DNA extracted from phages with better binding activity was sequenced and analyzed. Results After 4 times of screening, abundant specific phage model peptides were harvested. The results of ELISA showed that some phage model peptides had a fine binding activity. A total of 10 sequences related to promoting or inhibiting fibroblast cell proliferation were recognized. Conclusion TGF-β1-related phage model peptides can be harvested from a phage display 12-met peptide library, and some phage model peptides may inhibit keloid fibroblast proliferation.
