摘 要:探讨了纳米羟基磷灰石(nHAP)作用于大鼠肝线粒体时对线粒体酶活性的影响.将不同浓度的nHAP与线粒体直接作用,测定了线粒体标志酶琥珀酸脱氢酶(SDH)与超氧化物歧化酶(SOD)的比活性.结果显示:nHAP中水含量〈10%时,线粒体酶活性保持稳定;nHAP浓度≤0.56 g.L-1时,无吸光度值;在相同作用时间内,nHAP随浓度的增大,对线粒体SDH的比活性抑制作用增强;在不同作用时间(1~28 h)段,琥珀酸脱氢酶(SDH)的比活性较对照组有显著差异性(p〈0.05),组内两两相互比较有显著差异性(p〈0.01).nHAP对SOD比活性的影响随着时间的延长而增强,对SOD的比活性抑制作用随浓度的增加而增强,与对照组相比有显著差异性(p〈0.05).因此,nHAP对线粒体SDH及SOD比活性的抑制有剂量和时间的依赖性.This paper discussed the impact of nano-hydroxyapatite(nHAP) on the rat liver mitochondria activity.Through the direct reaction between different concentrations of nHAP and mitochondria,the activity of mitochondria succinate dehydrogenase(SDH) and the impact of nHAP on superoxide dismutase(SOD) activity were studied.The results show as follows: when the water content in nHAP is less than 10%,the mitochondria activity keeps steady;when the concentration of nHAP is less than or equal to 0.56 g·L-1,there is no absorbance;in the same reaction time,the higher the concentration of nHAP is,the greater inhibitory effect it has on the mitochondria SDH activity;in different reaction period(1-28 h),when compared with the control group,the activity of SDH has significant difference(p0.05),and when compared with each other in this group,there is also significantly great difference(p0.01).The impact of nHAP on SOD activity strengthens with the extension of time,and its inhibitory effect on SOD activity enhances with the increase of concentration,which has significant difference(p0.05) when compared with the control group.Therefore,the inhibitory effect of nHAP on SDH and SOD activity of mitochondria depends on the dose and the time.
