摘 要:目的建立HPLC测定大鼠血浆中的莫西沙星浓度的方法,并进行体内药动学研究。方法血浆样品采用二氯甲炕液一液萃取法提取。色谱柱为ZORBAXEdipseXDB—C8(150mm×4.6mm,5μm),流动相为水-0.1%三氟乙酸-乙腈(40:35:25),流速1.0mL·min-1,检测波长296nm,柱温30℃。结果莫西沙星浓度在0.05~10.00μg·mL-1内线性关系良好(r=0.9996);最低定量限为0.05μg·mL-1;低、中、高浓度的方法回收率为99.3%~106.0%,提取回收率为88.1%~94.1%:日内、日间精密度RSD均〈10%。在大鼠体内药动学过程符合二室模型。结论方法简便、经济、准确,可满足临床药动学研究的要求。OBJECTIVE To establish an HPLC method for the determination of moxifloxacin in rat plasma, and validated for pharmacokinetic study. METHODS The moxifloxacin was extracted from plasma by dichloromethane. Chromatographic separation was carried out on ZORBAX Edipse XDB-C8 column(150 mm×4.6 mm, 5 μm) with water-0.1% trifuoroacetic acid-acetonitrile(40 : 35 : 25) as the mobile phase at a flow rate of 1.0 mL.min-1. The detection wavelength was 296 nm and the column temperature was 30℃. RESULTS The linear range was 0.05-10.00 μg.mL-1(r=0.999 6). The lowest detectable limit was 0.05μg.mL-1. The recovery of the method was 99.3%-106.0%, and the extraction recovery was 88.1%-94.1%. RSDs of inter- and intra-day precisions were both less than 10%. The profile of moxifloxacin in vivo fitted a two-compartment model. CONCLUSION The method is simple, economic and accurate, which could be used for the pharmacokinetic study of moxifloxacin.
