摘  要:通过构建DNA池和测序的方法,分析了蓝塘猪和长白猪Mx1基因5’外显子1和内含子2共计1 514 bp(2 218-3 731)单核苷酸多态性。结果表明:在1 514 bp的序列上共发现了8处单核苷酸突变,1处为颠换,7处为转换,依次是:2 593G〉A、2 740T〉C、2 792C〉T、2 855C〉T、2 962C〉A、3 004C〉T、3 669T〉C、3 690A〉G。生物信息学预测发现猪Mx1基因内含子2上也存在多个转录因子结合位点,其中2 740T〉C和3 004C〉T分别位于潜在的转录因子LEF1和AP1的结合位点,但是没有位于结合位点的核心序列。The single nucleotide polymorphism of 5′ regulatory region(1 514 bp,2 218~3 731,exon2 and intron1) of Mx1 gene in Langtang and Landrace pigs was studied through DNA pooling and sequencing.The results showed that one transversion and seven transitions were found in the 5′ regulatory region of the gene.They were 2 593GA,2 740TC,2 792CT,2 855CT,2 962CA,3 004CT,3 669TC,and 3 690AG.The potential transcription factor binding sites were analyzed with the SNPs.The results by bioinformatics methods displayed that there were many potential transcription factor binding sites in the intron1 and exon2.The SNPs,2 740TC and 3 004CT were in LEF1 and AP1 binding sites.But the two ones did not exist in the core sequences of the two sites.
