摘  要:采用辣椒抗病基因同源序列(Resistance gene analogues)(RGA2)设计1对特异引物,对高抗辣椒疫病品种CM334和极感疫病品种Early Calwonder(EC)进行多态性扩增,开发出共显性的RGA-STS1200标记。利用STS1200标记在具有不同疫霉菌生理小种抗性的辣椒材料CM334、PBC459、PBC137-1和对辣椒疫霉菌3个生理小种都不具抗性的辣椒材料EC、茄门和B2间进行多态性扩增。结果表明,由3个辣椒疫病抗性材料所扩增出的条带大小一致,而由3个感辣椒疫病品种所扩增出的条带大小一致。说明STS1200标记可用于鉴定辣椒疫病抗、感材料。同时,利用共显性的RGA-STS1200标记在苗期及时剔除CM334和EC正反杂交所得F1代假杂种,进一步构建用于辣椒疫病抗性研究用的F2代分离群体。这是少数报道利用RGA-STS标记对辣椒疫病抗性进行鉴定,也是第一次利用RGA-STS标记对作物种子真伪进行鉴定。We developed a pair of primers based on the full-length disease resistance gene analogue(RGA2) cloned and released by our research group.Polymorphic application was conducted between CM334,an extremely resistant accession to Phytophthora capsici and Early Calwonder(EC),a highly susceptible accession to Phytophthora capsici,and obtain a co-dominant RGA-STS1200 marker.We checked the polymorphism of the RGA-STS1200 marker in three pepper accessions,CM334,PBC459 and PBC137-1,which conferred resistance to different Phytophthora capsici races,as well three completely susceptible species EC,Qiemen and B2.Results showed that the three resistance accessions possess the same band and the three susceptible species own the other band.This indicated that the STS1200 marker can be used to distinguish resistant or susceptible pepper species to Phytophthora capsici.Furthermore,we removed the false hybrids of F1 derived from CM334 and EC in the seeding stage with the assistant of co-dominant RGA-STS1200 marker and constructed a F2 segregation population used for the research of resistance to Phytophthora capsici.This is few STS marker developed from RGA of pepper that used to distinguish the pepper resistant to Phytophthora capsici.This is also the first report demonstrating the utility of RGA-STS markers for the assessment of genetic purity of hybrids in crop plants.
