摘 要:[目的]筛选出一种树脂,并优化大孔吸附树脂纯化金樱根总黄酮成分的工艺。[方法]比较ADS-7、HPD300、AB-8、D-101型4种大孔吸附树脂对金樱根粗总黄酮提取液的吸附-解吸效果,考察上样液浓度、洗脱液浓度对解吸效果的影响。[结果]AB-8型大孔树脂对金樱根总黄酮成分吸附分离效果最佳;其纯化金樱根最佳条件为以20.0 ml浓度为0.200 mg/ml的样液进行吸附,用4 BV 50%乙醇进行洗脱。最佳工艺下,金樱根总黄酮的精制度为205.40%。[结论]通过该研究得到的方法可获得高纯度的金樱根总黄酮。[Objective] To obtain a suitable macroporous resin,and to optimize its purification process for total flavonoids of root of Rosa laevigata Michx..[Method] The adsorption and desorption effect of four resins like ADS-7,HPD300,AB-8 and D-101 were comparatively studied and effect of concentration of loading solution and eluent concentration were investigated.[Result] AB-8 resin was the optimum purification resin for total flavonoids of root of Rosa laevigata Michx.,and the optimum purification conditions were:loading concentration of 0.200 mg/ml,loading volume of 20.0 ml,eluent of 50% ethanol of 4 BV.Under the optimal conditions,the purification rate of total flavonoids of root of Rosa laevigata Michx.reached as high as 205.40%.[Conclusion] The high quality total flavonoids from root of Rosa laevigata Michx.was obtained in this work.
