摘 要:以不同发育时期和不同部位的菠萝果实为材料,比较了改良SDS法、改良CTAB法和Trizol试剂盒法提取RNA的效果。结果表明:3种方法提取的RNA差异明显,其中改良SDS法的效果最佳,其提取的RNA质量较好,无DNA污染,OD260/OD280和OD260/OD230接近2.0,产量达到464.3μg/g.FW以上,该方法提取的RNA能满足RT-PCR、Northern blot和cDNA文库建立等分子生物学研究的需要。Modified SDS,modified CTAB and Trizol Kit were used to extract total RNA from pineapple fruits of different sections and development stages.The results showed that there were significant difference among the RNA isolated by using this three isolation methods,and the optimizing modified SDS was considered to be the most favorable methods for RNA isolation from different sections.RNA with high yield and quality,and had not DNA contamination could be obtained,Both the ratios of OD260 to OD280 and OD260 to OD230 were approximately to 2.0,meanwhile,the yield of RNA was 464.3 μg/g·FW.RNA isolated by this method could be used to further study of RT-PCR,Northern blot,cDNA library construction and other biotechnological researchers.
