摘 要:采用PCR-RFLP方法,对红色原鸡、杏花鸡、隐性白洛克鸡和丝羽乌骨鸡的IGF1R基因的18个位点的遗传多样性进行研究。结果表明,18个位点的基因频率在这4个群体中的分布差异很大,绝大部分的SNP最小等位基因频率均大于5%;独立χ2分析显示,除了位点G17445596A、T17416994G、A17313488G和C17337042G,其他14个位点的基因型分布在不同鸡品种间存在显著或极显著差异(P〈0.05和P〈0.01);红色原鸡、杏花鸡、隐性白洛克鸡和丝羽乌骨鸡4个群体之间的平均多样性差异均不显著。单倍型分析发现,在所研究的区域,红色原鸡、杏花鸡、隐性白洛克鸡、丝羽乌骨鸡等在鸡IGF1R基因内分别划分为5、4、4、3个单倍型块;每个块内2~3个单倍型就可以代表整个块90%以上的遗传信息。此外,还发现鸡IGF1R基因中9个SNP位点可能与复杂经济性状有关,这为以后在经济性状上对该基因的进一步研究提供了参考依据。Eighteen variation sites in the chicken IGF1R gene were genotyped using PCR-RFLP method to study genetic diversities of four chicken breeds including Red jungle fowl(RJF),Xinghua(XH),Silkies(S),White Recessive Rock Broliers(WRR).The results indicated that the allele frequencies of 14 sites were significant different among the four chicken populations,moreover,minor allele frequencies of most of SNP were higher than 5%.The chi-square test revealed that there were significant differences of allele frequencies among four chicken populations(P0.05 and P0.01) for all sites except for G17445596A,T17416994G,A17313488G,and C17337042G.The mean diversity of RJF,XH,S,and WRR were 0.354±0.155,0.407±0.141,0.401±0.163,0.373±0.136,respectively.However,there was not significant difference between any two breeds.Based on the four-gametal ruler,the analyzed region of RJF,XH,S,and WRR could be divided into 5 disruptive haploblocks,4 disruptive haploblocks,4 disruptive haploblocks,and 3 disruptive haploblocks,respectively.Generally,2 to 3 haplotypes within each block could represent for over 90% diversity information.This study predicted that 9 SNP might be associated with complicated economic traits,and provided original information for elucidating the possible effects of IGF1R gene on chicken economic traits.
