多囊卵巢综合征患者子宫内膜“植入窗”期凋亡基因的表达及定位研究

摘　　要：目的探讨多囊卵巢综合征(Polycystic ovarian syndrome,PCOS)患者子宫内膜"植入窗"期凋亡相关基因的差异性,并对部分基因进行了定位研究。方法将PCOS患者和对照组各6例共分为3组,分别取"植入窗"期的子宫内膜组织,应用基因芯片技术,检测其差异表达基因,对芯片数据进行处理和生物信息学分析,得到了差异表达的凋亡基因,并对下调的Fas相关死亡结构域蛋白(Fas-associated death domainprotein,FADD)基因运用反转录-PCR及免疫组织化学方法对进行了表达及定位研究。结果 PCOS 3组中均明显差异表达的基因有194条,有9条为凋亡相关基因,其中5个上调,4个下调;免疫组织化学研究发现,FADD免疫阳性物质位于PCOS患者"植入窗"期子宫内膜腺上皮细胞的细胞浆内。结论 PCOS患者"植入窗"期子宫内膜部分凋亡基因表达紊乱,是影响其植入率的原因之一。Objective To investigate the apoptotisapoptosis genes differential expression in endometrium of ＂window of implantation＂ in polycystic ovary syndrome（PCOS）.Methods Twelve women were enrolled and divided into three groups respectively,in which six were normal women,the other were PCOS patients.The tissuees of endometrium of ＂window of implantation＂ of twelve cases were curettaged from the PCOS groups and the control groups.By using gene chips,we investigated the differential expressed apoptotisapoptosis genes of tissuees of endometrium of ＂window of implantation＂ in PCOS patients.The data was processed and analyzed using bioinformatics knowledge.By using RT-PCR and immunohistochemistry,we also investigated the expression and distribution of FADD（Fas-associated death domain protein） in endometrium of ＂window of implantation＂ of PCOS,which is one kind of down-regulated apoptosis gene in PCOS.Results As compared with control group,there are 194 diferentially expressed gene,including 9 genes associated with apoptosis,in which 5 genes were up-regulated,and 4 genes were down-regulated in PCOS patients.Endometrial epithelioglandular cells in PCOS showed FADD immunoreactivity,which were located in cytoplasm with negative nuclei.Conclusion The apoptotisapoptosis genes of endometrium of ＂window of implantation＂ in PCOS patients are disordered.This might indicate a declined capicity of implantation in the window time in PCOS patients.
