摘  要:目的:建立液相色谱-串联质谱法,用于测定人血浆中倍他米松(BOH)的质量浓度。方法:以二丙酸倍氯米松为内标(IS),采用Waters XTerra C18柱(2.1mm×100mm,3.0μm)为固定相、甲醇-水(88:12,V/V)为流动相;流速为0.15mL/min;质谱采用电喷雾离子源,多反应离子监测模式,BOH和IS的检测离子对分别为m/z361.1→m/z307.0和m/z543→m/z433.1。结果:血浆中BOH质量浓度在0.05-20μg/L范围内与峰面积比值线性关系良好(r2=0.9995),最低定量浓度为0.05wg/L,日内、日间精密度(RSD)均小于12.0%,相对回收率均在98.3—103.4%范国内。结论:谊方法具有较高的准确度,线性范围宽,方法灵敏,专一性好,操作简便,适用于BOH血药浓度的测定及临床药代动力学研究。Objective To determine the concentration of betamethasone in human plasma by liquid chromatographic tandem mass spectrometric method. Methods Beelomethasone dipropionate was used internal standard. The evaluation of betamethasone was carried out by Waters XTerra column (2.1 mm × 100 mm, 3. 0 μm) with the mobile phase of methanol / water ( 88 : 12, V/V ) ,and the ions of analyte were detected by multiple ionic reaction monitoring mode ( MRM ), the mass transition pairs of m/z 361.1→m/z 307.0 and m/z 543.0→m/z 433.1 were used to detect betamethasone and IS, respectively. The flow rate was 0.15 mL/min. Results The calibration curve for plasma betamethasone was linear in range of 0.05 - 20μg,/L( r = 0. 999 5 ). The detection limit of betamethasone was 0. 051μg/L. Within - day RSD and between - day RSD were less than 12.0%, the relative recovery were all within 98.3 - 103.4%. Conclusion This method is accurate,sensitlve,specific and convenient,which could be used in the determination of plasma betamethasone and its pharmacokinetic study in clinic.
