摘 要:采用细乳液聚合法制备了嗜硫磁性纳米微球,磁球粒径较小,具有灵敏的磁响应性。经嗜硫性配基(2-巯基噻唑啉)修饰后,在pH=4.0的低浓度柠檬酸缓冲液中可特异性吸附免疫球蛋白G,并实现了在较低盐浓度下被吸附抗体的有效脱附,在吸附过程中表现出明显的单分子层吸附过程。在分离过程中几乎不损伤抗体的生物活性,所分离抗体的生物活性高于99%。磁性微球的平衡吸附量为53.04mg/g,相比于微米级磁性微球的平衡吸附量(3.1mg/g),提高了17倍。Nano-sized magnetic polymer spheres with intense magnetic response were synthesized by miniemulsion polymerization. After modified by 2-mercaptothiazoline on the surface of these magnetic nanospheres, these magnetic particles exhibited a strong specificity towards IgG. It was found that the qe (53. 04mg/g) of magnetic polyaner nanospheres increased by 17 times compared with the qmax (3.1 mg/g) of micro-spheres. This specific binding of IgG could be performed at low citric acid concentration(pH=4.0) and the responding desorption was performed at low salt concentration. Thus, not only the selective recognition of antibodies could be carried out under physiological conditions, but also the efficient elution could be achieved under mild conditions. It was much favorable to preserve the bioactivity of the antibodies and to increase the purity of IgG. The bioactivity of the isolated antibodies all exceeded 99% .
