摘  要:为获得皖西白鹅PIT-1基因全部编码区序列,探明该基因在皖西白鹅早期发育阶段的表达规律,该研究分别以成年皖西白鹅基因组DNA和垂体总RNA为模板进行PCR和RT-PCR扩增、测序;采用荧光定量PCR技术分别分析皖西白鹅PIT-1基因在0周、2周、4周、7周、10周、15周和30周龄的相对表达量。结果获得了1 014bp的cDNA序列(GenBank accession number.FJ389720),其中编码区的长度为1 011 bp。同源分析结果显示:获得的编码区序列与其他物种的同源性均在60.0%以上,与鸭的同源性高达97.2%,所编码的氨基酸序列与禽类其他物种的同源性在86.0%以上。荧光定量PCR分析结果表明:初生时PIT-1基因在垂体内即有一定水平的表达;4周龄前性别间基因的表达模式存在差异;7周龄后性别间的表达模式趋于一致,表达量下降;到10周龄以后又开始上升,并在15周龄以后趋于稳定。This study was conducted to obtain the whole coding domain sequence(CDS) of PIT-1 gene in Wanxi white goose and to explore its expression in goose early growth stages.PCR,RT-PCR and sequencing technologies were employed,with Wanxi white goose genomic DNA and pituitary total RNA as the templates.A cDNA sequence of 1 014 bp was acquired,and the length of CDS was 1 011 bp.The nucleotide similarity of CDS of goose PIT-1 gene with other species was higher than 60.0%,and a high degree of homology with duck(97.2%) was exhibited.The similarity of putative amino acids sequence with other poultry species was more than 86.0%.Fluorescence real-time PCR technology showed that the expression of PIT-1 gene was kept at a certain level right after birth in both genders,and differed in genders before 4-week age.The expression declined after 7 weeks,then increased from 10-week age,and kept stable after 15-week age.
