摘  要:为探索辣椒黄瓜花叶病毒(CMV)的抗性相关基因,采用mRNA差异显示技术探讨抗、感辣椒在接种CMV前后基因表达的差异。对获得的87个差异片断进行回收、重扩增和克隆后共获得26个阳性克隆。数据库比对分析结果表明,仅有7个cDNA克隆在GenBank中找到了相似的功能,与能量代谢、转录因子和代谢酶有关,其余cDNA片断在数据库中未发现匹配信息;抗、感病辣椒材料均具有诱导表达特点,抗病材料(Vc16a)差异表达条带的数目比感病材料(SS69)少。研究结果为进一步分离和克隆辣椒抗CMV相关基因提供了试验依据。To find the gene related to cucumber mosaic virus(CMV)-resistance in pepper,RNAs isolated from pepper materials with resistance and sensitivity to CMV were used to study the differentially expressed genes before and after inoculation by mRNA differential display technique.The results showed 87 differential display bands.Among them,26 positive clones were selected by recovering,second amplification and cloning.Only seven fragments were found homologous sequences in GenBank database.These sequences mainly participated in ATP syntheses,transcript factors and metablic enzyme;there were still other fragments with no hit in the GenBank.The characteristic of induction expression in the two tested materials was shown as well.The number of differentially expressed bands in resistant material was less than that in susceptive material.
