摘 要:目的研究大环内酯类药物对哮喘大鼠肺组织窖蛋白-1(Cav-1)和凋亡相关蛋白B细胞淋巴瘤/白血病-2(Bcl-2)蛋白表达的影响及二者的相互关系。方法将40只大鼠随机分为4组:对照组、模型组、红霉素干预组(A1组)、红霉素+地塞米松干预组(A2组)。以卵白蛋白致敏制作大鼠哮喘模型。应用免疫组织化学法和蛋白印迹方法检测不同组别大鼠肺组织Cav-1和Bcl-2蛋白的表达。结果免疫组织化学法和蛋白印迹方法检测不同组别大鼠肺组织Cav-1和Bcl-2蛋白表达结果一致。模型组大鼠肺组织Cav-1蛋白表达显著低于其他各组(Pa<0.01)。A2组大鼠肺组织Cav-1蛋白表达显著高于A1组(P<0.05),但A1组和A2组均显著低于对照组(Pa<0.01)。模型组大鼠肺组织Bcl-2蛋白表达显著高于对照组和A2组(P<0.05,0.01),但与A1组相比差异无统计学意义(P>0.05)。A1组、A2组与对照组相比,Bcl-2蛋白表达均显著升高(Pa<0.01),但A1组与A2组比较差异无统计学意义(P>0.05)。Cav-1与Bcl-2蛋白表达呈显著负相关(r=-0.618,P<0.01)。结论大环内酯类药物可协同糖皮质激素通过上调Cav-1表达和下调Bcl-2表达促进细胞凋亡,从而发挥抗炎作用。Objective To investigate the effects of macrolides on the expressions of caveolin-1(Cav-1) and B cell lymphoma/leukemia-2(Bcl-2) proteins in the lung tissue of rats with asthma,and to explore the relationship between Cav-1 and Bcl-2 protein expression. Methods Asthmatic rat models were established by repeated ovalbumin sensitization,40 cases of Sprague-Dawley rats were divided into 4 groups randomly,including control group,model group,erythromycin interference group(group A1) and erythromycin + dexamethasone interference group(group A2).Expressions of Cav-1 and Bcl-2 proteins in lung tissues of rats in 4 group were detected by immunohistochemistry and Western blot. Results Expressions of Cav-1 and Bcl-2 proteins in lung tissues measured by immunohistochemistry were consistent with those measured by Western blot in every corresponding group.Expression of Cav-1 protein in model group was significantly lower than that in other groups(Pa0.01).Cav-1 protein in the group A2 was significantly higher than that in group A1(P0.05);while that in both group A1 and group A2 were significantly lower than that in control group(Pa0.01).Expression of Bcl-2 protein in model group was significantly higher than that in control group and group A2(P0.05,0.01),while there was no significant difference of expression of Bcl-2 protein between model group and group A1(P0.05).Significant differences of Bcl-2 protein were observed between the group A1 and control group,between the group A2 and control group,respectively(Pa0.01).There was no significant difference of Bcl-2 protein between the group A1 and group A2(P0.05).Negative correlation was observed between the expression of Cav-1 and Bcl-2 protein(r=-0.618,P0.01). Conclusions Macrolides combined with glucocorticoids may play an antiinflammatory role by up-regulating Cav-1 expression and down-regulating Bcl-2 expression in the lung tissues,which can promote cell apoptosis.
