摘  要:目的观察灵芝多糖和虫草多糖组成的复方制剂(LCF)对小鼠酒精性肝损伤的保护作用和对2.2.15细胞分泌乙型肝炎病毒表面抗原(HBsAg)和乙型肝炎病毒e抗原(HBeAg)的影响。方法末次给液20 min后,后4组一次性给予50%乙醇20 ml,禁食24 h后处理,在酒精性肝损伤模型中,小鼠随机分成对照组,模型组和LCF低中高剂量组,分别灌胃双蒸水和对应试液(等容量)3 d,测定血清ALT和AST的活性,肝匀浆MDA、GSH和TG的含量;在2.2.15细胞模型中,LCF稀释成不同浓度加入2.2.15细胞进行培养,每3 d换含同浓度药物培养液,分别收集3 d、6 d以及停药后3 d的培养液,测定HBsAg和HBeAg的含量。结果与模型组比较,LCF高剂量组能显著降低血清中ALT和AST的水平,低中剂量组显著升高肝匀浆中GSH含量和降低TG含量,中高剂量显著降低肝匀浆中MDA含量。LCF连续给予3 d时1和2 mg.ml-1剂量组、6 d时0.25、0.5、1和2 mg/ml剂量组及停LCF3 d的0.5、1.0和2.0 mg/ml剂量组分别能显著抑制HBsAg的分泌。各剂量组在各测定时间点对HBeAg均无显著影响。结论 LCF对酒精性肝损伤具有保护作用,对2.2.15细胞分泌HBsAg具有显著抑制作用。Objective To study the protective effect of the compound preparation of Ganoderma lucidum polysaccharide and Cordyceps polysaccharide(LCF) on alcoholic liver injury in mice and the effect of LCF on hepatitis B surface antigen(HBsAg) and hepatitis B e antigen(HBeAg) secretion in 2.2.15 cell.Method In alcoholic liver injury model,the mice were randomly divided into control group,model group and low-,middle-,high-dose groups of LCF(80,200, 400 mg/kg).After oral treatment for 3 d,the levels of ALT and AST in serum,the contents of MDA,GSH and TG in liver homogenate were measured.In 2.2.15 cells treated with LCF,which were diluted into 0.25,0.5,1.0,and 2.0 mg/ml and the medium of the same concentration was replaced every 3 d.The fluid was collected on 3,6 and 3d after treatment.The contents of HBsAg and HBeAg were detected.Results Compared with the alcoholic liver injury model group,LCF 400 mg/kg could significantly decrease the levels of ALT and AST,while LCF 80 and 200 mg/kg could significantly increase the content of GSH and reduce the content of TG.LCF 200 and 400 mg/kg,could significantly diminish the content of MDA.In 2.2.15 cell model,LCF signiticantly suppressed 2.215 cell to secrete HbsAg in 1 and 2 mg/ml group after 3 d administration,in 0.25,0.5,1 and 2 mg/ml group after 6d adimistration,and in 0.5,1 and 2 mg/ml group after 3d cessation following 6d administration.There was no significant effect of LCF on HBeAg secretion.Conclusion LCF has significant protective effect on alcohol-induced liver injury and pronounced inhibitory action on HBsAg secretion in 2.2.15 cell.
