摘 要:目的研究纤维蛋白黏合剂的潜在免疫原性。方法建立间接ELISA法检测抗体条件;新西兰白兔创伤实验及Balb/c小鼠实验考察纤维蛋白黏合剂的免疫原性。结果新西兰白兔创伤实验中,给药一周后兔体内即可检测出抗体,给药2周后抗体水平明显升高,6周后开始下降。各组血清滴度均呈现先上升后下降的趋势;小鼠实验中,纤维蛋白黏合剂作用于小鼠后,高、中、低各剂量组均能够激活小鼠的免疫系统,增强嗜中性粒细胞的吞噬功能;增加小鼠免疫器官重量及脾细胞的转化能力。结论研究表明,纤维蛋白黏合剂对于兔诱发特异性抗体,对小鼠表现出明显的免疫原性。This study aims to investigate the potential immunogenicity of fibrin sealant.Firstly,indirect ELISA was used detect specific antibodies in sera,and the immunogenicity of fibrin sealant and fibrinogen were investigated by means of the injury experiment in New Zealand rabbits and the medication experiment in Balb/c mice.We found that in the case of injury experiment in New Zealand rabbits,the specific antibodies appeared after one week,significantly rose up after 2 weeks,and dropped 6 weeks later,with dynamic tendency of serum titer rising and later dropping in every group.In the medication experiment on Balb/c mice,fibrin glue could motivate the immune system in Balb/c mice in high,middle and low dose groups,enhance the effect of neutrophil and showed a proliferative ability of splenocytes in mice,and fibrin glue could also enhance the weight of immune organs.These results conclude that fibrinogen in the main components of fibrin sealant could induce specific antibodies in rabbits and show apparent immunogenicity in mice.
