摘  要:目的:考察老鹳草提取物对免疫性肝损伤小鼠的保护作用。方法:分别采用卡介苗(BCG)+脂多糖(LPS)、刀豆蛋白A(ConA)诱导建立小鼠免疫性肝损伤模型,灌胃给予老鹳草提取物(100,200,400mg.kg-1),观察小鼠血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)和乳酸脱氢酶(LDH)的活性及血清总蛋白(TP)、白蛋白(ALB)、细胞因子γ干扰素(IFN-γ)和白细胞介素4(IL-4)含量变化;检测肝组织匀浆中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;进行肝脏病理组织检查。结果:不同剂量老鹳草提取物均可明显降低小鼠血清中AST,ALT活性及肝组织匀浆MDA含量(P<0.05或0.01),并能使降低的SOD活性升高;中、高剂量老鹳草提取物(200,400 mg.kg-1)可不同程度降低血清中IFN-γ和IL-4含量及LDH活性(P<0.05)。不同剂量老鹳草提取物可使血清蛋白含量维持正常比例,肝组织损伤均有不同程度减轻。结论:老鹳草提取物对小鼠免疫性肝损伤具有一定的保护作用。Objective:To investigate the protective effect of the extracts from Herba geranium on immunological liver injury in mice.Methods:The immunological liver injury in mice was induced by BCG+LPS and ConA,respectively.The serum levels of AST,ALT and LDH and the concentrations of TP,ALB,IFN-γ and IL-4 were measured.The liver was isolated for pathohistological examination;SOD activity and MDA content in liver homogenates were determined.Results:Herba geranium extracts(100,200 and 400mg·kg-1,ig) significantly decreased the serum levels of ALT and AST,and the contents of MDA in liver homogenates(P0.01 or 0.05);and increased the activity of SOD in liver tissue.Herba geranium extracts(200 and 400mg·kg-1,ig) decreased the serum concentrations of IFN-γ and IL-4,and the serum level of LDH(P0.05).The concentration of total protein in serum was continuously in a normal state.Pathohistological examination showed that Herba geranium extracts effectively decreased degeneration and necrosis of liver cells.Conclusion:The extracts from Herba geranium have a protective effect on immunological liver injury in mice.
