摘 要:目的:探讨抗原抗体反应检验若干人肿瘤细胞分泌β-hCG情况,和抗hCGCP22抗血清体外抑制肿瘤的作用。方法:采用细胞免疫荧光染色测定5个肿瘤细胞株是否表达β-hCG,用CCK-8试剂盒检测CP22抗血清(AS CP22)和β-hCG羧基端37肽(CTP)抗血清(AS CTP)体外抑制肿瘤细胞生长的光密度。结果:以AS CP22为探针和AS CTP为阳性对照,体外培养的人胰腺癌BXPC-3、人卵巢癌HO-8910和人结肠癌Lovo细胞膜上都能观察到绿色荧光,提示肿瘤细胞均表达β-hCG亚单位,而人前列腺癌PC-3和Du-145细胞则未检测到绿色荧光;与正常兔血清(NS)比较,AS CP22和AS CTP均可显著的体外抑制BXPC-3和HO-8910细胞生长,P<0.001。AS CP22抑制BXPC-3和HO-8910肿瘤细胞的作用都强于AS CTP,P<0.05。结论:人胰腺癌BXPC-3、人卵巢癌HO-8910和人结肠癌Lovo细胞在体外培养的情况下均表达β-hCG亚单位,且它们的体外生长都能被兔抗hCG嵌合肽(CP22)抗血清体外抑制。OBJECTIVE: To observe whether several human tumor cells secret β-hCG subunit and the serum against hCG chim- eric peptide (CP22) inhibits tumor cell proliferation in vitro. METHODS: Whether five strains of human tumor cells express β-hCG were determined using immunofluorescence staining, and the optical density of tumor cells growth inhibited by anti-CP22 an- tiserum (AS CP22) and anti- β-hCG C-terminal 37 residue synthet- ic peptide serum (AS CTP) in vitro was determined using CKK-8 kit. RESULTS.. Using AS CP22 as probe and AS CTP as positive control, the green immunofluorescence on human pancreatic (BX- PC-3), ovarian (HO-8910) and colorectal (Lovo) cells were found in vitro, suggesting that these tumor cells expressed β-hCG subunit, whereas that did not observed on prostate tumor (PC-3 and Du-145) cells. Compared with normal rabbit serum, the growth of BXPC-3 and HO-8910 cells in vitro could be inhibited notably by rabbit AS CP22 and AS CTP (NS, P〈0. 001). Also, the inhibi- tory effects of AS CP22 to BXPC-3 and HO-8910 cells were better than those of AS CTP (P〈0.05) when using four different dilutions of both rabbit antisera. CONCLUSION: In five strains of human tumor cells selected in this study, the BXPC-3, HO-8910 and Lovo cells express β-hCG subunit in vitro and their growth in vitro can be inhibited by the rabbit serum to hCG CP22.
