摘 要:目的探讨大鼠矽肺模型不同染尘剂量与血清硅元素水平、克拉拉细胞蛋白(CC16)和表面活性蛋白D(SP-D)之间的剂量-反应关系并进行比较。方法 145只雄性W istar大鼠按不同染尘剂量(10、30、50、70 g/L)及不同时间点(染尘后1、3、5、7、14、21和28 d)分为对照组及28个染尘组。取大鼠血液及肺组织,采用电感耦合等离子体质谱仪(ICP-MS)检测矽肺模型大鼠血清中硅元素水平,酶联免疫吸附测定法(ELISA法)定量测定大鼠血清中CC16和SP-D水平。结果随着染尘剂量的增加和时间的延长,模型组大鼠血清硅元素的水平呈现逐渐增高的趋势;CC16水平于造模后即开始降低,自第7天开始与对照组比较,差异有统计学意义(P<0.05),并随着时间的延长和染尘剂量的增加水平不断减少;SP-D水平于造模后即开始增加,与对照组比较,在第5天差异有统计学意义(P<0.05),在第14天最高,21 d起开始逐渐下降。结论矽肺模型大鼠血清硅元素水平在早期低剂量染尘时即明显升高,且早于CC16和SP-D的变化,三者水平均随染尘剂量和时间的不同而变化,提示血清硅元素、CC16和SP-D水平变化可能为矽肺的早期诊断提供生物学指标,具有一定的意义及价值。Objective To explore the dose-response relationship of blood silicon,CC16 and SP-D in rats exposed to different doses of silica.Methods 145 male wistar rats were randomly divided into a control group and 28 treated groups according to the SiO2 doses(10,30,50,70 g/L)and different time points(1,3,5,7,14,21 and 28 days).The levels of serum silicon were determined by inductively coupled plasma mass spectrometry(ICP-MS),while CC16 and SP-D were tested by ELISA.Lung tissues were stained by hematoxylin and eosin,and observed by microscope.Results The levels of blood silicon increased gradually with the increase of dust doses and time.The level of serum CC16 in the 7th day was significantly lower than that of the control group(P0.05),and it decreased gradually with dust doses and time.The levels of serum SP-D in the 5th day was significantly higher than that of the control(P0.05),and peaked at the 14th day.Conclusion Change of serum silicon came earlier than the expression of CC16 and SP-D did,and the levels of these indexes changed with dust doses and time.There were some value and meaning in the early diagnosis and pathogenesis for silicosis by determining the levels of serum silica,CC16 and SP-D.
