摘  要:目的观察人BAFF-R-IgG4mut融合蛋白对BXSB狼疮鼠CD4+T淋巴细胞增殖活性的影响。方法体外分离、纯化BXSB狼疮鼠脾脏CD4+T细胞,流式细胞技术检测CD4+T细胞BAFF-R表达,MTT法观察不同剂量BAFF-R-IgG4mut融合蛋白对BAFF刺激的CD4+T细胞增殖活性的影响。结果台盼蓝染色分离后的BXSB狼疮鼠脾脏CD4+T细胞活性率为(93.6±3.2)%;流式细胞技术检测BXSB狼疮鼠脾脏CD4+T细胞表达BAFF-R;100ng/ml、200ng/ml、400ng/ml BAFF-R-IgG4mut蛋白作用于BAFF和CD3抗体刺激的CD4+T细胞后,CD4+T细胞增殖抑制率分别是56.24%、67.07%和75.45%,较单纯BAFF+CD3抗体组差异有统计学意义(P<0.05)。结论 BAFF-R-IgG4mut融合蛋白可体外抑制BXSB狼疮鼠CD4+T细胞增殖活性。Objective Study the effect of human BAFF-R-IgG4mut fusion protein on proliferative activity of CD4+T lymphocytes in murine BXSB lupus.Methods Isolation and purification of BXSB lupus mouse spleen CD4+T cells.BAFF-R expression in CD4+T cells was detected by flow cytometry.The effect of different doses of BAFF-R-IgG4mut fusion protein on the proliferative activity of CD4+T cell under BAFF and CD3 antibody stimulation were assessed by MTT.Results Activity of CD4+T cells separatiing from BXSB lupus mouse were(93.6±3.2)% through detection by Trypan blue staining.Spleen CD4+T cells in BXSB lupus mice could express BAFF-R.After 100ng/ml,200ng/ml,400ng/ml BAFF-R-IgG4mut protein stimulating the CD4+T cells,the CD4+T cell prolliferative inhibition rate were 56.24%,67.07% and 75.45% respectively which were significant different than that of BAFF+CD3 antibody group(P0.05).Conclusions BAFF-R-IgG4mut fusion protein inhibited proliferative activity of CD4+T cell in BXSB lupus mice in vitro.
