摘 要:比较不同条件下聚酰胺树脂对甘草黄酮的静态吸附和动态吸附的效果,确定甘草黄酮分离纯化的最佳条件。结果表明,聚酰胺树脂层析纯化甘草黄酮的最佳条件是:上样液浓度为3.51 mg/mL,pH 10.0,用70%乙醇以1.00 mL/min流速洗脱。对从1 g甘草中提取得到的甘草黄酮采用层析柱层析,发现回收率和纯度都较高。在最佳条件下,经两次层析可使甘草黄酮纯度达到49.75%。The optimum experimental conditions was determined by comparing the features of static and dynamic absorption of crude flavonoids under different conditions.The results showed that the optimum experimental conditionds were as follows: loading concentration of flavonoids was 3.51 mg/mL,pH 10.0,and the most effective desorption was obtained with 70% of alcohol as eluent with a flowing rate of 1.00 mL/min.The highest recovery rate and purity were achieved when use a chromatography column with inner diameter of 1.5 cm and height of 16 cm for the amount of licorice flavonoids extracted from 1 g Glycyrrhiza.Under optimum conditions,the content of flavonoids reached 49.75% after purified by polyamide resin adsorption chromatography for two times.
