双波长RP-HPLC法测定清开灵胶囊中栀子苷和黄芩苷的含量

摘　　要：目的建立测定清开灵胶囊黄芩苷和栀子苷含量的RP-HPLC法。方法色谱柱：Hibor C18键合硅胶柱（4.6 mm×160 mm,5μm）。流动相：A相为0.2%磷酸水溶液,B相为乙腈。采用梯度洗脱,流动相：0-10min：B相-A相（12∶88）,流速0.8 mL/min;10 min之后：B相-A相（25∶75）,流速：1.0 mL/min。检测波长：240 nm（栀子苷）、278 nm（黄芩苷）。柱温：30℃。结果栀子苷在0.16-2.4μg范围内线性关系良好,平均加样回收率为96.94%,RSD=2.08%（n=6）,黄芩苷在0.18-2.7μg范围内线性关系良好,平均加样回收率为100.18%,RSD=2.01%（n=6）。结论本法简便、快速、灵敏,精密度和重现性良好,专属性强,准确度高,可作为该制剂有效成分的含量测定控制方法。Objective To establish a RP-HPLC method for determination of geniposide and baicalin content in qingkailing capsule.Methods A Hibor C18 was used（4.6 mm×160 mm,5 μm）and detective wavelength was 240 nm（geniposide）,278 nm（baicalin）.The mobile phase：acetonitrile-0.2% phosphoric acid（12∶88）（0～10 min）,the flow rate was 0.8 mL/min;acetonitrile-0.2% phosphoric acid（25∶75）（after 10 min）,the flow rate was 1.0 mL/min.Results The standard curve of geniposide was linear over the range of 0.16-2.4 μg（r=0.999 7）,the standard curve of baicalin was linear over the range of 0.18-2.7 μg（r=0.999 9）.The average rate of geniposide recovery was 96.94%,and RSD was 2.08%,the average of baicalin recovery was 100.18%,and RSD was 2.01%.Conclusion This method is reliable,accurate and suitable for the determination of geniposide and baicalin in qingkailing capsule.
