摘  要:目的:建立超高效亲水性液相色谱与串联四级杆飞行时间质谱仪联用技术(UPLC HILIC/Q-TOF-MS)对毛茛黄酮类成分进行分析的方法。方法:用ACQUITY UPLC BEH HILIC色谱柱,以甲醇∶水=1∶1含10mM乙酸铵(A)/乙腈∶甲醇=95∶5含10mM乙酸铵(B)梯度洗脱,使用ESI离子源,正、负离子模式下采集数据。结果:根据元素组成分析、二级质谱和相关文献,鉴定、推断出毛茛中11个主要成分,分别是小麦黄素,牡荆素,荭草素,异荭草素,芹菜素-6-C-β-D-葡萄糖苷-8-C-α-L-阿拉伯糖苷,小麦黄素-7-O-β-D-葡萄糖苷,芦丁等。结论:经过超高效亲水性液相色谱的分离,借助Q-TOF-MS测定的准确分子量及二级质谱信息可以判定毛茛中的主要黄酮类成分,为毛茛黄酮类成分分析提供了一种快速有效的方法。Objective:To investigate the flavonoids in Ranunculus japonicus by Ultra-performance Hydrophilic interaction liquid chromatography and tandem quadrupole time-of-flight mass spectrometry technique(UPLC HILIC /Q-TOF-MS).Methods:The chromatographic separation was performed on a ACQUITY UPLC HILIC C18 column with a gradient elution of methanol∶water=1∶1 containing 10mM ammonium acetate(A) and acetonitrile∶methanol=95∶5 containing 10mM ammonium acetate(B).The data were collected with the positive and negative ion mode,using ESI ion source.Results:Eleven compounds were deduced by elemental compositions,MS/MS fragment ions,the comparison with standard compound as well as literatures.There were tricin,vitexin,orientin,isorientin,6-C-β-D-glucosyl-8-C-α-L-arabinosylapigenin,tricin-7-O-β-D-glucopyranosy,rutin and etc.Conclusion:Using positive ion and negative ion information,the major constituents were identified by HILIC separation,Q-TOF-MS accurate determination of the molecular weight and MS/MS data.This is a rapid and effective method for identification of the flavonoids in Ranunculus japonicus.
