中药复方君子汤对白血病细胞株K562/VCR耐药性的逆转作用

摘　　要：目的观察中药复方君子汤(FFJZ)对白血病耐药细胞株K562/VCR细胞耐药性的逆转效果。方法采用MTT法、流式细胞仪术分别检测FFJZ、环孢霉素A(CsA)和维拉帕米(VRP)作用于白血病耐药细胞株K562/VCR的耐药指数、逆转指数及细胞内ADM的浓度,与CsA、VRP进行比较。结果 FFJZ、CsA和VRP在体外对白血病K562/VCR细胞耐药性均有明显的逆转作用(P<0.01),FFJZ对白血病K562/VCR细胞的逆转指数与CsA和VRP相似,能显著提高K562/VCR细胞对阿霉素的敏感性,在有效浓度范围下对细胞本身无明显毒性作用。结论 FFJZ可能成为治疗白血病多药耐药安全有效的逆转药物。Objective To investigate the reversal effect of Chinese medicine compound FFJZ on the multidrug resistance of human leukemia K562/VCR cell line.Methods The resistance index and reversal index were detected by MTT assay,and the intracellular Adriamycin（ADM） was detected by flow cytometry after K562/VCR cell line were interfered by FFJZ,cyclosporine A（CsA） and verapamil（VRP） respectively.Results FFJZ could significantly reverse the drug-resistance of K562/VCR cells（P0.01）,and its reversal effect on the multidrug resistance of human leukemia K562/VCR cell line was similar to that of CsA and VRP.FFJZ could increase the sensitivity of K562/VCR cells to ADM,and had not the toxic effect on the K562/VCR cells in effective dose.Conclusion FFJZ may become a safe and effective multidrug resistance-reversing agent with low toxicity in leukemia chemotherapy.
