摘 要:[目的]建立松针中莽草酸测定与有机溶剂提取分离方法。[方法]以水为试剂提取分离马尾松松针中莽草酸;用NH2色谱柱,乙腈和2%H3PO4(90:10,V:V),柱温25℃,流速1ml/min,检测波长213nm,高效液相色谱(HPLC)法测定莽草酸含量。[结果]所得样品供试液中莽草酸杂质少;线性范围为0.29~2.88μg(R2=0.9999),平均回收率为98.30%,RSD为0.74%(n=5)。[结论]提取分离方法高效、环保、成本低;HPLC法适用于松针及其他天然植物中莽草酸的测定;马尾松松针中莽草酸平均含量为1.14%。[Objective]The determination of shikmic acid in pine needle and organic solvent extraction and separation method were established.[Method]The shikimic acid in leaf of pine needle was extracted and separated and its content was determined with the method of high performance liquid chromatography(HPLC) with the NH2 being taken as separation column;acetonitrile-2% H3P04(90:10),mobile phase;the temperature of column,25 ℃;the flowing velocity,1 ml/min;and the testing wavelength,213 nm.[Results]The sample of shikimic acid extracted was with less impurity.The linear range of the shikimic acid was in 0.29-2.88 μg(R2 =0.999 9),the average collection was 98.30%,RSD was 0.74%(n =5).[Conclusion]The extraction and separation method was suitable for the determination of shikimic acid with high performance and environmental protection and low cost.The average content of shikimic acid in leaf of pine needle was 1.14%.
