脂氧素A4对子痫前期患者脐静脉内皮细胞分泌内皮素-1的影响

摘　　要：目的:研究脂氧素A4(lipoxinA4,LXA4)对子痫前期患者脐静脉内皮细胞分泌内皮素-1(endothelin-1,ET-1)的影响。方法:取子痫前期患者和正常孕妇组脐带及脐静脉血清,分离并培养脐静脉内皮细胞(human umbilical veinendothelial cell,HUVEC),分为正常细胞对照组、子痫前期细胞组和LXA4药物干预组(1 nmol/L,10 nmol/L,100 nmol/L)。RT-PCR检测各组细胞ET-1mRNA水平;酶联免疫吸附试验检测各组细胞上清液中ET-1的分泌水平。结果:LXA4降低子痫前期患者HUVEC内ET-1mRNA基因表达水平(P<0.05),而对正常细胞组无影响(P>0.05);LXA4明显抑制子痫前期脐静脉内皮细胞分泌ET-1(P<0.01),对正常细胞亦无影响(P>0.05)。结论:LXA4抑制子痫前期脐静脉内皮细胞ET-1 mRNA表达及分泌,为临床应用LXA4治疗妊娠期高血压疾病提供重要的实验依据。Objective：To study the effect of lipoxin A4 on the secretion of endothelin-1 in endothelial cells of umbilical vein in patients with preeclampsia. Methods：The umbilical cord samples and serum samples of umbilical vein were obtained from patients with preeclampsia and normal pregnant women,then endothelial cells of umbilical vein were isolated,cultured and divided into normal cell control group,preeclampsia cell group and lipoxin A4 intervention group（1 nmol/L,10 nmol/L,100 nmol/L）.RT-PCR was applied to detect the levels of endothelin-1 mRNA in different groups;ELISA was used to detect the levels of endothelin-1 in supernatant liquid of different groups. Results：Lipoxin A4 reduced the expression level of endothelin-1 mRNA in endothelial cells of umbilical vein in preeclampsia cell group（P0.05）,but it had no impact on the expression level of endothelin-1 mRNA in endothelial cells of umbilical vein in normal cell control group（P0.05）;lipoxin A4 inhibited the secretion of endothelin-1 in endothelial cells of umbilical vein in preeclampsia cell group（P0.01）,but it had no impact on the secretion of endothelin-1 in endothelial cells of umbilical vein in normal cell control group（P0.05）. Conclusion：Lipoxin A4 inhibits the expression and secretion of endothelin-1 mRNA in endothelial cells of umbilical vein in patients with preeclampsia,which lays an important experimental foundation for clinical application of lipoxin A4 in treatment of hypertensive disorder complicating pregnancy.
